In recent years, there are some disturbing questions have been emerged in ourcountry’s food safety area and the ecological environment field. Such as milkpowder in the " melamine", pork in "lean meat powder", Jiangsu Taihu Lake "bluealgae events", ConocoPhillips "bohai sea oil leakage events"............. These kinds ofquestions just like current global outbreak of financial crisis.It swept over the wholeearth, and influence seriously the people’s life and health, more affecting everyone’sheart. So, everybody pay attention to an common issue at the present,which proceeda series of effective detection to the food and environment.Determining the colonies number is an important and basic work of the qualitydetection with the food,air and water quality analysis. For example, the total numberof colonies in the air is an important index to test atmospheric pollution degree,andthis is also a standard to evaluate contaminated degree in the water. At present, thedomestic usually put method of ordinary nutrition agar pour plate as a nationalstandards inspection. However, because of this method requires large quantities ofsample to test, and the program is complicated, time-consuming, lowefficiency.Meanwhile count rely solely on the macroscopic observation that has somesubjective and uncertainty,bad reproducibility, big error.So it needs improvement.These years, some overseas company also have developed a set of colonies automaticcounting system.however, because of its equipment complex,for the size of thecolonies and form of waiting for inspection have limited, it failed to realizeminiaturization and fast detection.With digital imaging technology and the development of computer technology,based on an image acquisition and image analysis processing images automaticanalysis system came into being. Because of it can rapid, automatic and objectiveanalysis with the characteristics of image, improve the accuracy of analysis, andreduce the workload.Becoming a research hotspot at home and abroad in recent years.This paper adopts the752H x582V resolution of CCD camera, ARM chipprocessor, LED lighting system, test paper set up the system hardware platform ofcolonies. Among them, the CCD camera uses SONY480line of1/3. CCD trigger andARM chip adopt S3C2410chip of production by Beijing Universal PioneeringTechnology Co.,LTD. Lighting system from using LED light of multi-angle. The testpaper of colony adopts the production with the total number of rapid test colony ofGuangzhou LaiTe high-tech industrial company.In order to improve the precision of the image processing, we first proceed imagepreprocessing to the collected colonies image. Pretreatment process normally include digital, geometric transformation, normalization, smooth, rehabilitation and enhancesteps, etc. For make the image become simple and reduce the amount of data, also canhighlight the outline of the colonies, the image after pretreatment get binary imagewith gray weighted threshold algorithm of binary processing. Using Houghttransformation remove edge image of colony test paper, and using distancetransformation combined with watershed algorithm segment sticky colonies.Withlabel method of eight neighborhood to label the colony, finally complete countcolonies. Realized the objectivity of the colony counts and quickness.Through the experimental testing, the system can be completed the colony countsof daily collection.It compared with the actual visual count, there were some errors,but the error basically controlled at5%. For daily colony detection has the certainauxiliary function.
