In this study, we investigated the programmed cell death characteristics of ulmus pumila L.seeds which were experienced6days control deterioration in37℃and100%relative humidity(RH),including nuclear shrinking, reactive oxygen species bursting and caspase-3-like activity.The methods what we used including DAPI, TUNEL, DCF staning conbined with laser scanning microscopy (CLSM) and caspase activity kit.In order to obitain the changing of Ca2+flux follows by seed aging, Non-invasive Micro-test Technique(NMT) was choiced. The results indicated that the condensation of nuclear chromatin was gradiant apearing by the aging time. The apoptosis was firstly observed in the epidermal cells,later in cotyledons and hypocotyls.Our research also showed that the distinct spatial-temporal ROS signature was convinced with other hallmarks of PCD(programmed cell death) which gradually increased at5d.Otherwise,the caspase-3-like activity was found during5d control deterioration which play a key role in seed aging.And the NMT results showed that the apparent Ca2+influx increased before the2d(1200pmol cm-2s-1) and decreased from the3d to5d. Our data indicate that the ROS, Ca2+influx and caspase-3-like also act as PCD mediators in the process induced by deterioration.
