Eualiopsis binata, identified as a new apomictic species, is of great interest for its ability to fix heterosis. By Screening through the library, we get microsatellite primers. Genetic diversity of Eulaliopsis binata populations and lines were analyzed with SSR primers. If we could examine the F1 progeny plants by SSR primer, we maybe get the molecular markers tightly linked to apomixes and different reproduction plants.The results are as follows:1. Therefore, we constructed a microsatellite-enriched library. By Screening through the library, we get 70 pairs of microsatellite primers. After the PCR test, we found that 27 pairs sueeessfully amplified Polymorphic bands.2. Genetic diversity of 11 Eulaliopsis binata populations and 71 Eulaliopsis binata lines were analyzed with 8 SSR primers.123. alleles was detected and the average number of alleles was 15.375.6~34 polymorphic fragments were amplified by each SSR marker;These loci exhibited high levels of polymorphic information content,0.762~0.953(average 0.872); Genetic dissimilarity coefficients ranged from 0.70 to 0.97. These 11 populations of Eulaliopsis binata could be clustered into 4 groups. Xi-chuan, Dan-jiang, Yun-xian, Heng-yang, Yun-xi, Nei-xiang, Xi-xia, San-yang were clustered into one group. However, Xing-zhi, Bai-she, Yang-xian showing a high genetic diversity, were clustered into other 3 groups. By UPGMA analysis,71 Eulaliopsis binata lines could be divided into 3 groups, and results were consistent with Eulaliopsis binata group classification.3. The analyses for plants from hybrid progenies of Xi-xia population were employed using screened polymorphism SSR primers. A F1 progeny plants of Xi-xia3 exhibit the bands of both parents. The offspring is a true hybrid. The parent of the plant may be sexual reproduction.4. Using leaves of seeding as control, populations were examined by Flow Cytometry. Finally, individual plants of Xi-xia3 had two peaks 2C:3C, the others had three peaks 2C:3C:4C. So Xi-xia3 might have sexual reproduction.
