Taste cell-based sensor is a kind of cell-based sensor, which using taste receptor cells (TRCs) as recognition elements, and it can determine the existence or the concentration of a certain tastant (such as sweet, bitter, salt, sour tastants). The sensor works in real-time, rapidly, lossless, thus attains certainly qualitative and quantitative analysis.Based on principle, structure and characteristics of cell-based sensor, a taste cell-based sensor system was proposed here. It utilizes carbon screenprinted electrode (CSPE) for base, which using TRCs as recognition elements, with electrical cell-substrate impedance sensing (ECIS) for examining method, with bistable stochastic resonance (BSR) for data treatment. Finally it carried out NCI-H716cells-based sensor to recognize the sweet tastants, and STC-1cells-based sensor to recognize the bitter tastants. The results are shown as follows:(1) Taste cell-based sensor experimental system designThe system includes CSPE selection and characterization, and automatic perfusion system, and taste cell observe system, and electrical impedance spectrum system, and BSR treating data system; and at the same time designing the method of TRCs cultured on CSPE surface. The system test expresses it contented the experiment request, and the optimal TRCs seeding concentration was determined by impedance spectrum; and TRCs liveses rate is up to above90%.(2) The study of NCI-H716cells-based sensor to recognize sweet tastantsFirst of all, the taste receptor protein T1R2and T1R3of sweet and a-gustducin about taste signal transduction were expreesed in the NCI-H716cells demonstrated by immunohistochernistry. Secondly, the culture method of NCI-H716cells on the surface of CSPE was proved by scanning electron microscope (SEM) attribute. Finally, NCI-F1716cells-based sensor can recognize natural or artifical sweeteners with same sweet intensity, and recognize distinctly the sucrose in the mixed solution with the different basic tastants, at the same time, the concentrations of sweeteners were differentiated by different MAX of signal-to-noise ratio (SNR) and noise intersity.(3) The study of STC-1cells-based sensor to recognize bitter tastantsFirstly, the taste receptor protein T2R38of bitter and a-gustducin about taste signal transduction were expreesed in the STC-1cells demonstrated by immunohistochemistry. Secondly, the culture method of STC-1cells on the surface of CSPE was proved by SEM attribute. Finally, STC-1cells-based sensor can distinguish bitter tastants, and distinguish distinctly the quinine in the mixed solution with the different basic tastants, at the same time, the concentrations of bitter tastants were recognized by different MAX of signal-to-noise ratio (SNR) and noise intersity.
