The hypoglycemic effects of traditional Chinese medicines compounded shenmaikeli (SMKL) were studied in diabetic mice induced by alloxan (ALX), diabetic rats induced by streptozotocin (STZ), insulin-resistant mice induced by hydrocortisone sodium succinate (HCSS), hyperglycemic mice induced by adrenaline (Adr) and glucose (Glu). Furthermore, the mechanisms of hypoglycemic of SMKL were investigated.SMKL (0.5, 1.0, 2.0g/kg, ig) showed manifest effect on reducing the hyperglycemia in ALX-induced diabetic mice. At the same dosage, SMKL could significantly antagonize the hyperglycemia induced by Adr and Glu. And it could also antagonize the effect of insulin-resistance induced by HCSS. At the same time, SMKL could obviously improve the decrease of body weight, the increase of drinking water volume and urine volume and death rate in mice.SMKL (0.35, 0.7, 1.4g/kg, ig) showed manifest effect on reducing the hyperglycemia in STZ-induced diabetes rats, it could significantly enhance the contents of insulin in serum and liver glycogen in normal rats. In the model rats induced by STZ, the level of NO in serum, SOD in plasma, GHb were increased. Meanwhile, the level of T-CHO, TG in serum, MDA in plasma, excretion of urine albumin, and MDA in liver and pancreas decreased. At the same time, SMKL could obviously improve the decrease of body weight, the increase of urine volume and the death rate in rats. In pathological observation, SMKL could significantly antagonize pancreasβ-cell and renal lesion.The results demonstrated that the hypoglycemic effect of SMKL was associated with the reparation of pancreasβ-cell and the stimulation on secretion of insulin. It could promote the synthesis of liver glycogen and the usage on glucose in tissue or inhibit the decomposition of liver glycogen. At the same time, it could improve the tolerance of glucose and the insulin-resistance in the diabetic models. Furthermore, SMKL also had a strong precaution on the diabetes complications including vascular, nervous system, renal, and so on, by improving fat metabolism, eliminating free radicals, antagonizing oxidative stress and renal lesion.
