Effects Of Aldosterone On Gap Junction Connexin43Expression In H9c2Cardiomyocytes

Objective：To investigate the effects of different concentrations of aldosteron onthe expression of Cx43protein and mRNA in H9C2cells in order to make clear theprotective mechanism of arrhymias。Methods：1. H9C2cells were cultured with Dulbecco's Modifed Eagle Medium of Highglucose,then were randomly divided into control group and experimental group,theexeperimental group was exposed to different concentrations of aldosterone(1x10^-8mol/L,1x10^-6mol/L,1x10^-4mol/L),and control group in not contain tire of bovineserum DMEM medium for24hours。2. After24hours,we carried out RT-PCR analysis to show the change ofcontent of Cx43mRNA in different concentrations of aldosteron groupes。3. After24hours,we carried out Western blot analysis to show the change ofcontent of Cx43protein in different concentrations of aldosteron groupes。Result：1. Different concentration of aldosterone group compared with the controlgroup,1x10^-8mol/L aldosterone groupe's expression of Cx43mRNA was increased(P <0.05), there is significant difference,and1x10^-6mol/L,1x10^-4mol/L aldosteronegroupe's expression of Cx43mRNA has no obvious difference(P＞0.05)。2. Different concentration of aldosterone group compared with the controlgroup,1x10^-8mol/L aldosterone groupe's expression of Cx43protein was increased (P<0.05), and1x10^-6mol/L,1x10^-4mol/L aldosterone groupe's expression of cx43protein was decreased (P <0.05), there is significant difference。Conclusion：The study found that the effect of different concentration of aldosterone onH9C2cell's Cx43protein and mRNA is different. Low concentration(1x10^-8mol/L)aldosterone group's Cx43protein and mRNA is increased,and high concentration(1x10^-6mol/L,1x10^-4mol/L)aldosterone group's Cx43protein is decreased,but Cx43mRNA has no obvious change.So speculate that aldosterone may be involved ingap junction remodeling through its dual effect on the expression of Cx43,thusleading to the development of arrythmias。...
