Objective:To explore the influence of s-phase kinase associated protein2(SKP2)expression and its significance in lung adenocarcinoma A549cells after the treatmentof Pemetrexed(PMX).Methods:According to the concentration of PMX, we set one control group andfour experimental groups:0μM(group A),0.01μM(group B),0.1μM(group C),1μM(group D),10μM(group E). The growth of A549cells handled by differentconcentration PMX for24h,48h,72h,was tested by MTT assay; The effect ofdifferent concentration PMX handling for48h on the cell cycle phase distribution ofA549cells was analyzed by flow cytometry; The expression of SKP2protein of A549cells handled by different concentration PMX for48h was detected by Western blot.Results:MTT assay showed that PMX could significantly inhibit the growth ofA549cells, and the highest inhibition ratio was (78.61±3.66)%at the concentrationof10μM when A549cells were cultured for72h (P<0.01). Meanwhile, s-phase cellsof group C, D, E were more than group A(P<0.05,P<0.01,P<0.01), and the numberof s-phase cells was increasing with the concertration of PMX. The difference ofSKP2protein expression among each group was significantly(P<0.01), andcompared with control group SKP2protein expression significantly increased inexperimental groups(P<0.01). That increasing was obvious with the increasing ofPMX in certain concentration range.Conclusion:PMX could inhibit the growth of A549cells and arrested them ins-phase. The expression of SKP2protein in PMX-treated A549cells was upregulated,which might be concerned with the chemotherapy effect of PMX.
