The neuroscience research of human brain began to develop and applied to the clinical treatment,after the human brain stereotaxic methods were found. The research of human's brain nuclei, from the globus pallidus, ventral lateral thalamic nucleus to the amygdala, the lenticular nucleus and caudate nucleus,was deeper and deeper. The brain stem is the vital location of the many nerve nucleus. However, at present,three-dimensional morphology, structure and size of each nucleus in the stem, are poorly understood are poorly understood. Three-dimensional morphology and position of gracile nucleus has been explorated in this study as the beginning of the brain stem nucleus research.Two cases of Chinese adult men were selected as the brain stem sample. After the different advantages and disadvantages of a variety of slicing methods were balanced, the paraffin method of making slices was choosed. The point was well marked before specimen of the brain stem was cut into several pieces. Then the wax block was made and slicing began.There were many kinds of the brain nerval tissue staining methods. Cresyl violet staining was determined. Slices, using cresyl violet staining dye, were put under the microscope, according to the location point.The next step was image correction, whose purpose was to prepare for later image segmentation. Image segmentation, the necessary process of computer three-dimensional reconstruction, is to extract the interested information. Combining the automatic segmentation and manual segmentation to image segmentation was adopted by this research. Canny edge detection method for gracile nucleus in this study was used as the automatic segmentation method in the automatic segmentation.Three-dimensional reconstruction method includes surface reconstruction and volume reconstruction. Compared the advantages and disadvantages of the two methods, this research was oriented to intensive data field surface reconstruction method for gracile nucleus three-dimensional reconstruction.The nuclei of three-dimensional measurement and statistical methodological exist some disadvantage. The shape of an average-Measurement of mean shape index of gracile nucleus three-dimensional reconstruction was adopted in this paper. There were three results in this research.First, gracile nucleus was the irregular nuclei, whose three-dimensional average shape was symmetrical nuclei. But gracile nucleus was slightly larger on the right than on the left. Three-dimensional reconstruction of gracile nucleus can be observed at any angle and cut with arbitrary orientation. Three-dimensional morphology and the position of Gracile nucleus were intuitive, vivid image on the show.Second,this research showed that on the right side of gracile nucleus,length width height and volume were8.659mm,5.842mm,4.897mm and67.80mm3. On the right side of gracile nucleus, length width height and volume were8.411mm,4.250mm,3.560mm and51.36mm3. Third, this study provided a continuous atlas of gracile nucleus.
