Objective:The research object is Codonopsis thalictrifolia Wall.. The purpose is to observe and test the effects of water extraction from Codonpsis thalictrifolia Wall.on the reproductive system of female SD rats. The effect of drug serum on granulose cells and its mechanism were researched by the method of Serum pharmacology. This study will provide scientific data for clinical usage of Codonpsis thalictrifolia Wall.to cure reproductive disorder.Method:Female rats were administrated with water extraction of Codonpsis thalictrifolia Wall, or Codonopsis pilosula Nannf.. After2weeks, rats'weight of ovary and uterus were tested. Estradiol and progesterone of rats were tested by radioimmunoassay. FSH, LH, GSH, SOD, and MDA were tested by ELISA.The method of granulosa cells cultured in vitro was established. The effect of drug serum on granulosa cells was researched by the method of serum pharmacology. Its mechanism was researched by blocking the pathway of cAMP, Ca2+and protein synthesis.Result:After having been fed the water extract from Codonopsis thalictrifolia Wall. or Codonopsis pilosula Nannf. for14days, it can be found that ovary weight and MDA content in serum have been reduced in female rats(P<0.05). The content of FSH and GSH in serum has been significantly increased (P<0.05).The activity of granulosa cells was tested by trypan blue exclusion experiment and the content of estradiol in granulose cells. The result told us the best condition of granulosa cells cultured in vitro is the5%concentration of drug serum, and24h culture time.The estradiol secretion of granulosa cells was reduced by drug serum (p<0.05). But the content of MTT and3β-HSD, the secretion of progesterone of granulose cells kept the same with control (P>0.05). H89could resist the effect of Codonopsis thalictrifolia Wall. on granulose cells(p<0.05).Conclusions:Water extraction from Codonpsis thalictrifolia Wall. could reduce freeradical in rats by increasing serum GSH and decreasing serum MDA.Serum of Codonpsis thalictrifolia Wall, could reduce the estradiol secretion of granulosa cells, but not effecting the productivity and differentiation of granulose cells.Serum of Codonpsis thalictrifolia Wall, could effect the estradiol secretion of granulose cells by cAMP pathway.
