Objective:Observed Rhodopsin kinase expression changes in the different duration of type2diabetic rat retinal tissue and positioning of expression, to explore its possible mechanism in the process of diabetic retinopathy, and to provide the experimental basis for further in-depth study of the pathogenesis of diabetic retinopathy.Methods:Select the experimental group of GK rats32and a control group of normal rats32and divided into four groups according to duration of diabetes, diabetes4w group,8w,16w group,24w group (n=8). Control group and experimental group, weeks of age, the relative should be grouped (n=8). Using fundus fluorescein angiography (fundus fluorescein angiography, FFA), the retinal changes observed in normal rats and the different course of diabetic rat model in vivo. Technology, using real-time fluorescence quantitative polymerase chain reaction (Real time-PCR), analysis of Rhodopsin kinase factor in the different duration of diabetes on rat retinal tissue expression, and immunohistochemical detection Rhodopsin kinase factor in the retinal tissue of diabetic rats expression and location.Results:1,FA examination results showed that:the overall incidence of approximately82.6%. GK rats after4weeks of age can be found in the fundus changes and course of the disease occurred more than eight weeks to greatly increase the risk of retinal changes.2,Real time-PCR results showed that, Rhodopsin kinase expression changes in the control rats was no significant difference (P>0.05). Compared with the control group, diabetic group of its expression is decreased, and as the birds grew to express more and more low, began to decrease when the4w,24w when dropped to the lowest (P <0.01).3,Immunohistochemistry results showed that the positive expression of Rhodopsin kinase in rat retinal rod cone cell layer, outer nuclear layer, outer plexiform layer; control rats of expression was no significant difference (P>0.05). Diabetic rats4w4w compared with normal control group, positive strength began to decline,8w and4w compared to decline further, with the progression of positive intensity of expression is becoming less and less,24w lowest (P<0.01).Conclusion:1,s the disease increases, the incidence of DR gradually increased, but the duration of disease, the key factor is not the DR and decisions lesions. Week-old rats of different diabetes blood glucose levels and the degree of DR is not an exact match.2,hodopsin kinase gene expression changes may be involved in the development of neural retinal lesions in diabetic rat model. Neural retinal lesions early in type2diabetes can occur.3,hodopsin kinase abnormal gene expression caused by photoelectric conduction disturbances may be one of the causes of diabetic retinopathy early occurrence of visual dysfunction.
