[Objective]:To study the effect of Panax Notoginsenoside(PNS) treatment on human bile duct carcinoma cell (QBC939) proliferation inhibition.apoptosis. cell cycle. migration and invasion.[Methods]:We used MTT assay to measure cell viability of QBC939cell line at different PNS concentrations and time intervals to determine the optimum PNS treatment effectiveness. Under these experimental conditions.we also examine the cell morphological changes using the microscope. We used flow cytometry to analyze QBC939cell apoptosis and cell cycle under the PNS treatment. We used cell scratch test to investigate cell migration and invasion.[Results]:In vitro culturing condition.we observed more than95%QBC939cells alive. The cells reached the logarithmic grow phase after12hour of culturing. The MTT test was performed after48hours cell inoculation, and it showed time and dose dependent inhibition effects of the PNS on QBC939cell line. PI-Annexin V flow staining assay showed obvious apoptotic cell ratio increased and cell cycle was blocked at G2/M phase as the PNS concentration increased. The transwell and cell scratch test showed that the PNS could prohibit cell migration and invasion.[Conclusion]:Our study showed that PNS treatment can prohibit QBC939cell proliferation in a dose and time dependent manner. We also found that the PNS could induce cell apoptosis.modify cell cycle and reduce cell migration and invasion. Therefore.we postulate that the PNS could be a potential treatment drug in treating human bile duct carcinoma.
