ObjectiveThrough the research of different grades of glioma tissue in ERCC1, IDH1and normal brain tissue expression, discussion of the ERCC1and IDH1expression in the relationship of brain glioma, clarify, development and molecular mechanism, in order to further discuss the prognosis of gliomas assessment and chemotherapy regimens, the results may serve as a clinical brain glioma aneurysms can provide certain basis.MethodsBy S-P immunohistochemical technique, were examined in42patients with different levels of brain glioma and10cases of normal human brain tissue ERCC1and IDH1expression, and were statistically correlated analysis.Results1.18patients with gliomas of42were positive with IDH1. The overall positive expression rate is42.85%; The positive rate of expression of different levels as follows: the positive rate of grade I glioma was36.37%(4/11); The positive rate of Grade Ⅱ glioma was40%(4/10); The positive rate of Grade Ⅲ glioma was50.00%(6/12);The positive rate of grade IV was44.44%(4/9). IDH1expression rate increased with the rise of Glioma pathological level.By statistical analysis, the different of different levels of glioma was no statistically significant,(P>0.05)2.22patients with gliomas of42were positive with ERCC1. The overall positive expression rate is52.38%. The positive rate of expression of different levels as follows: the positive rate of grade I glioma was27.27%in (3/11);The positive rate of grade Ⅱ glioma was40.00%(4/10);The positive rate of grade Ⅲ glioma was66.67%(8/12) The positive rate of grade Ⅳ was77.78%(7/9).ERCC1expression rate increased with the rise of Glioma pathological level.By statistical analysis,the different of different levels of glioma existing statistical significance.3.10cases of cerebral hemorrhage in the resection line decompression surgery within the normal brain tissues of IDH1and ERCC1expression was negative or weakly positive. The expression of IDH1and ERCClwere significantly different(P<0.05) compare with glioma tissue.4. There were no significant difference with gender and age of the expression of IDH1and ERCC1in the42patients'glioma organization.Conclusions1. different grades of glioma IDH1expression rate is different, and its rate of expression level increased with glioma and assumes the trend of escalation, glioma higher level of IDH1expression rate is relatively higher, but statistically analyzes the differences were not statistically significant, IDH1expression and malignant degree of gliomas correlation.2. ERCC1expression in gliomas of different levels was of different, and its expression rate increased obviously with the rise level of gliomas, the different express exist statistical significance. The expression intensity was positively correlated with glioma grade, explain that IDH1expression was related with tumor grade.3. IDH1and ERCC1expression in glioma cells were correlated, suggesting two gene in glioma tumorigenesis, development, growth and malignant invasion process plays a reciprocal regulation, synergistic effect, can be used as a prognostic indicator and chemotherapy. On two kinds of detecting gene on glioma clinical treatment can play a certain reference role.4. The expression of IDH1and ERCC1in normal brain tissues was negative or weak positive.
