Part I GK rats fluorescein sodium fundus angiographyObjective:Examination of rat model of type2diabetes retinopathy incidence, severity and the duration of the relationship.Methods:Thirty-two spontaneous diabetic GK rats were chosen as experimental group and32age-matched normal wistar rats were selected as control group. According to the course of diabetes, the diabetic GK rats were further divided into4groups consisting of4weeks group,8weeks group,16weeks group, and24weeks group. Eight rats in each group. Those normal wistar rats were also further divided into4groups as the diabetic GK rats. Analysis of the rat model of retinal lesions and the course of disease by fundus fluorescein angiography (FFA) technology.Results:FFA examination results showed that GK rats DR occur not with the duration of diabetes and blood sugar level of positive correlation.Conclusion:Blood glucose levels and disease duration and not diabetic retinopathy severity and decided the key factor, but the course of the disease and the incidence of diabetic retinopathy was positively correlated. Part II Study on the gene expression and related effect about gene of Rpe65and Rod CNG Channel in rats diabetes modelObjective:To study the gene expression and location of Retinal pigment epithelium-specific protein65(Rpe65) and rod photoreceptor cGMP-gate channel(Rod CNG Channel)gene in diabetic rat retina, investigate the gene expression tendency and related effect of Rpe65and Rod CNG Channel gene in the course of diabetic rat retinopathy, provide theoretical basis for further research and early treatment of diabetic retinopathy.Methods:Thirty-two spontaneous diabetic GK rats were chosen as experimental group and32age-matched normal wistar rats were selected as control group. Compared with the normal control group, diabetic group of its expression is down, and with the course of the increase in expression was decreased (P<0.01). The difference was statistically significant2.Real-Time PCR results showed that the gene expression of Rod CNG Channel in control rats no significant difference (P>0.05). Compared with the normal control group, diabetic group of its expression is down, and with the course of the increase in expression was decreased (P<0.01). The difference was statistically significant.Conclusion:1. The gene expression changes of Rpe65and Rod CNG Channel may be involve in the development retinopathy events of type2dibetes in the rat model.2. The gene expression changes of Rpe65gene and Rod CNG Channel may be related in visual function dysfunction of early diabetes retina.
