| Objective Through testing the plasma's level of IFN-y, IL-10and IL-6in Chronic Hepatitis B patients, discussing the relationship between IFN-y, IL-10and IL-6level and Chronic Hepatitis B patients'condition. Providing some basis for the pathogenesis of infection and antiviral treatment to Hepatitis B.Methods Collecting119cases of Chronic Hepatitis B patients and17cases of health as the research object.119cases of Chronic Hepatitis B patients are divided into several groups according to the people's medical publishing house published 《infectious Disease》(6edition):Chronic mild-moderate Hepatitis B group(A) have75cases, Chronic severe Hepatitis B group(B) have12cases, Hepatitis B liver cirrhosis group(C) have20cases, Hepatitis B carriers group(D) have12cases.17cases of health as a control (E).Detecting the level of IFN-y, IL-10and IL-6with ELISA method in Chronic Hepatitis B patients and healthy controls'peripheral blood plasma.The level of IFN-y, IL-10and IL-6were compared among in various patient group, patient group and healthy controls respectively.We used fluorescent quantitative polymerase chain reaction(FQ-PCR) to detect serum HBV-DNA levels; used enzyme-linked imunnosorbent absorption method (ELISA) to detect HBV antigen and antibody. The results were statistical analysised by SPSS17.0so ftware.Results (1) The IFN-y level in Chronic severe Hepatitis B, Hepatitis B liver cirrhosis group. Hepatitis B carriers group are higher than the normal control group, there is statistical significance (B vs E,C vs E,D vs E, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005); The IFN-y level in Chronic severe Hepatitis B, Hepatitis B liver cirrhosis group are higher than the Chronic mild-moderate Hepatitis B group, there is statistical significance (B vs A,C vs A, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005); the IFN-y level in Chronic severe Hepatitis B group is higher than the Hepatitis B carriers group, there is statistical significance (B vs D, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005).(2) The IL-10level in Chronic mild-moderate Hepatitis B group, the Chronic severe Hepatitis B group, Hepatitis B liver cirrhosis group are higher than the normal control group, there is statistical significance (A vs E,B vs E,C vs E, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005); the IL-10level in Chronic severe Hepatitis B group is higher than Chronic mild-moderate Hepatitis B group, there is statistical significance (B vs A, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005).(3) The IL-6level in Chronic severe Hepatitis B, Hepatitis B liver cirrhosis group is higher than Chronic mild-moderate Hepatitis B group, there is statistical significance (B vs A,C vs A, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005); the IL-6level in Chronic severe Hepatitis B group is higher than Hepatitis B carriers group, there is statistical significance (B vs D, Kruskal-wallis Rank Test&Nemenyi Test, p<0.005).(4) The comparison of IFN-y, IL-10and IL-6's level between HBeAg (+)-CHB and HBeAg (-)-CHB, there is no statistical significance (Mann-whitney, p>0.05).(5) The IL-6's level of HBV-DNA (+) is higher than CHB HBV-DNA (-) CHB, there is statistically significance (Mann-whitney, p<0.05); while, the comparison of IFN-γ and IL-10's level between HBV-DNA (+) CHB and HBV-DNA (-) CHB, there is no statistical significance (Mann-whitney, p>0.05).(6) The IL-6level of no antiviral treatment is higher than antiviral treatment, there is statistical significance (Mann-whitney, p<0.05); while, the comparison of IFN-y and IL-10level between antiviral treatment and no antiviral treatment, there is no statistical significance (Mann-whitney, p>0.05).(7) The level of IL-6and IL-10in Chronic Hepatitis B patients'plasma were positively correlated (r=0.388, p=0.000); The level of IL-6and IFN-y in Chronic Hepatitis B patients'plasma were positively correlated (r=0.331, p=0.000); The level of IL-10and IFN-y in Chronic Hepatitis B patients'plasma were positively correlated (r=0.342, p=0.000)Conclusion (1) The level of IFN-y rise in Chronic Hepatitis B patients' plasma suggesting that Thi cells mediated celluar immune response may be involved in the organism for HBV removal.(2)The level of IL-10rise in Chronic Hepatitis B patients'plasma suggesting that IL-10may be involved in the body's own immune adjustment to restrain the strong cellular immune response.(3) The level of IL-6in the virus high copy group is higher than the virus low copy group in Chronic Hepatitis B patients'plasma; The IL-6level of no antiviral treatment is higher than antiviral treatment, suggesting that the level of IL-6may report HBV replication and the body's immune reaction degree.(4) The level of IFN-y. IL-10and IL-6in Chronic Hepatitis B patients'plasma rise as the degree of patient's condition increased, suggesting that they are involved in the body's immune response to the HBV virus.(5) IL-6and IL-10, IL-6and IFN-y, IL-10and IFN-y are all positively related in Chronic Hepatitis B patients'plasma, suggesting that they may have relastionship with the body's immune regulation of IFN-γ inhibit IL-6and IL-10, IL-10inhibit IL-6and IFN-γ's generation. make the body's immune to balance state. |
