Part1A Study of Conditions Influencing Polymerase Chain Reaction-Restriction Fragment Length Polymorphism in the Site of CCR5gene promoter region59029G/A PolymorphismObjective:To determine the optimum experimental conditions of59029G/A polymorphism in CCR5gene detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). Methods:The main influential factors were detected by PCR in serial concentration gradient and electrophoresis, the main factors were detected by RFLP in different systems. Results:(1) When denaturation temperature was94℃could achieved the clearest electropherogram.(2) When annealing temperature was58℃, PCR product level was higher than others and less non-specific products.(3) When primer concentration was0.4umol/L, PCR product level was as well as others but had less dimer of primers.(4) It was5U Bsp12861that could digest5μl PCR product in13μl digestion system. Conclution:The optimum conditions:denaturation temperature was94℃; annealing temperature was58℃primer concentration was0.4umol/L; the effective system was13μl including5μl DNA solution and5U enzyme. Part2A study of CCR5gene promoter region59029G/A polymorphism and its related factors for the Lincang regional Lahu ethnicObjective:To Investigate the correlation of the Lincang regions Lahu ethnic glucose and clinical factors. Methods:we choose1426cases of Lincang area Lahu individuals as the survey, grouped in accordance with blood glucose, gather relevant information such as height, weight, blood pressure and etc,.At the same time, we collect blood samples, urine samples to test relevant indicators, including liver and kidney function, blood glucose and lipids and etc. All the data to form a unified database, using the SPSS18.0statistical software to handle data, P<0.05sentenced significant difference. Expect to find clinically relevant factors that may affect blood glucose abnormalities in the development, severity and so on. Results:age. Part3A study of CCR5gene promoter region59029G/A polymorphism and abnormal glucose metabolism related factors for the Lincang regional Lahu ethnicObjective:To Investigate the correlation of the Lincang regions Lahu ethnic CCR5gene in the promoter region of the59029G/A polymorphism and clinical factors; Methods:The polymorphism of CCR5gene59029G/A polymorphism was detected by PCR-RFLP in1426unrelated Subjects. The genotype frequencies,allele frequencies and clinic indexs were compared among groups. Results:the risk of drinking group in HDL-C abnormal occurred is1.497fold than non-drinking group (P=0.002<0.05), the risk of Female HDL-C abnormal occurred is0.694times than men (P=0.006<0.05). the rest of the factors in the model was not statistically significant (P>0.05). Conclusion:(1) Lincang regions Lahu ethnic's CCR5gene promoter59029G/A polymorphism is exist;(2) Drinking may be Lincang Lahu ethnic's risk factors in HDL-C abnormal occurred;(3) the Lahu female's HDL-C abnormal is less than men;(4) We didn't found that Lincang area Lahu people CCR5gene59029G/A polymorphism associated with HDL-C.
