Objective:To detect the mutations and expression changes of isocitrate dehydrogenase (IDH1) and isocitrate dehydrogenase (IDH2) in colorectal neoplasms. And to explore its potential role for colorectal neoplasms diagnosis and treatment according to analyse the relationsip between the two genes and colorectal neoplasms.Methods:We detected the expression changes of IDH1and IDH2in111pairs of colorectal neoplasms and normal tissues with the technology of quantitative real time polymerase chain reaction (qRT-PCR). Meanwile we also dectected hot-spot mutations of IDH1and IDH2in308cases of colorectal neoplasms with the first generation of sequencing technology.Results:We did not detect the hot-spot mutations in308cases of colorectal neoplasms, which including IDH1Arg132, IDH2Arg140and Arg172. However, we detected mutations with another spot of IDH1p.I99M in colorectal neoplasm and normal tissue. The relative expression level of IDH1and IDH2are (0.082±0.175)and (0.136±0.280), respectively, in colorectal neoplasms. And the relative expression level respectively of IDH1and IDH2are (0.087±0.148) and (0.154±0.317) in normal tissues. There were no relationship between the IDH expression and clinical parameters (gender, age, differentiation degree, tumor diameter, lymph node metastasis, distant metastasis, distribution site) in colorental neoplasms.Conclusion:There were no association between the expression of IDH1and IDH2and the onset and development with colorectal neoplasms. And there were also no association between the expression of IDH1and IDH2and clinical parameters. Mutation IDH1p.I99M was detected in a very low rale.We think IDH1may be relevant with the occurrence and development in colorectal neoplasms. Whether it is to become a new target in the diagnosis and treatment with colorectal neoplasms need further study.
