Objective:To investigate the effects of Feixingqing Capsules in boleomycin-induced pulmonary fibrosis in rat and the expression of TGF-β1,Smad2/3in the lung, and it possible mechanism.Methods:Forty SPF male SD rats were randomly divided into5groups (n=8), the normal control group (CG),the model group(MG),the positive control prednisone group(DG),FXQ group(EG) and DT group (FG).Except those rats in normal control group, all rats were established pulmonary fibrosis by intra tracheal administration of bleomyci, CG were injected with equivalent dosage of saline. After the model of fibrosis was established for24h, the rats of DG, EG,FG were given the drug mentioned above respectively, while only saline was given in CG and MG. On day28day after treatment, each group were sacrificed respectively, and the lungs were harvest for conventional HE pathological obserbation. TGF-β1and Smad2/3protein expression in lung issues were detected by immunohistochemistry.Results:1. The lung tissue pathomorphology of rats in MG showed that typical pulmonary fibrosis process.Collagen fibers sediments increased obviously and the structures of pulmonary alveoli were damaged on28th day. The expression of TGF-β1and Smad2/3elevated and reached a high level.2. After early intervention to the rats with pulmonary fibrosis,the pathomorphology of rat lungs in DG,EG and FG showed pulmonary fibrosis variation further advanced.but the extent distinctly lower that MG. It were similar that lungs expression of TGF-β1and Smad2/3protein between MG,DG,EG and FG. All those data from three treated groups are higher than CG and lower than MG (P<0.05), EG and FG are lower than DG (P<0.05). and EG is lower than FG(P>0.05).Conclusions:1. TGF-β1and Smad2/3protein showed high level expression in bleomycin-indueded rats lung tissue, which suggested that TGF-β1and Smad2/3protein are important cytokine in the proceeding of pulmonary fibrosis, and some functions of TGF-β1implemented through Smad2/3protein.2. Feixingqing Capsules have a treatment effect on pulmonary fibrosis, possible mechanism is through inhibiting the expression of TGF-β1and Smad2/3in the lung of rats with pulmonary.3. Feixingqing Capsules had better intervention effect on pulmonary fibrosis, and compared with prednisone, the side effect was mild.
