| [Objective]:To observate the roles of hypocrellin extract HA (hypocrellin A) inhibit the Yunnan Xuanwei lung adenocarcinoma cell line XWLC-05in vitro,and further eluminate the possible mechanism of the effects of HA on XWLC-05cells.[Methods]:1.MTT (Methy thiazolyl tetrazolium) assay was used to evaluate the inhitory effects of HA on XWLC-05cells at different concentration and time.2. Cell morphologies were observed by invert microscope, fluorescence microscope,sanning electron microscope and transmission electron microscope to analize the apoptosis of XWLC-05cells treated with HA.3.FCM (the fluorescence flow cytomtry) DNA assay was applied to detect the cell cycle phase and Apoptosis process of XWLC-05cells treated witth HA.4. DNA ladder assay was applied to detect DNA fragment in XWLC-05cells treated witth HA.[Results]:1. HA could inhibit the growth of XWLC-05cells in time-and concentration-depedant manner.2. Obvious apoptotic morphology changes were observed by invert microscope, fluorescence microscope, sanning electron microscope and transmission electron microscope in XWLC-05cells treated with HA.3. FCM assay indicated that most of the cells were arrested in S phase and the apoptosis rate of the cells treated with HA was increased with the time span.4. DNA ladders were found in in XWLC-05cells treated with HA, which certified that HA inhibts XWLC-05cells proliferation by its promoting apoptosis roles.[Conclusion]:1. HA could inhibit the growth of the lung cancer cell line XWLC-05in vitro.2. The inhitory effect of HA on XWLC-05proliferation is probably through the way of inducting apoptosis. |
