Abstract/Summary:
[Objective] To study theinfluences of B(a)P upon the mitochondrionof BEAS-2B human bronchial epithelial cells.[Method] The experimental group was been culturedby DMEM cultivation liquid which contained B(a)P50umol/1,10umol/1,1.0umol/1,the control group was been cultured by normal DMEM cultivation liquid. When the cells were been cultured24h,48h,72h, using FCM etermination of mitochondrial membrane potential changes; using RT-PCR to detecte Results applying spssl7.0statistical software for statistical analysis.[Results]1. The changes of mitochondrial membrane potential. By the extension of culture time, the level of mitochondrial membrane potential of the experimental groups decreased gradually, but50umol/1B(a)P and10umol/1B(a)Pmitochondrial membrane potential downward trend is more pronounced (P<0.01).2. The relative expression quantity of Cyt-b mRNA. Cell culture after24h, the relative expression quantity of Cyt-b mRNA in50umol/1B(a)P,10umol/1B(a)P group increased,50umol/1B(a)P group a statistically significant difference (P<0.05),10umol/1B(a)P,1.0umol/1B(a)P groups'expression no difference between the control group (P>0.05).Cell culture after48h, The relative expression quantity of Cyt-b mRNA in50umol/1B(a)P,10umol/1B(a)P group increased, has significant statistically significant difference (P<0.01),1.0umol/1B(a)P groups'expression no difference between the control group (P>0.05).Cell culture72h, The relative expression quantity of Cyt-b mRNA in50umol/1B(a)P,10umol/1B(a)P group reduced,50umol/1B(a)P group a statistically significant difference (P <0.05),10umol/1B(a)P,1.0umol/1B(a)P groups'expression no difference between the control group (P>0.05) [Conclusion] B(a)P can damage the mitochondria of BEAS-2B.The higher the concentration and the longer duration of action, the effect of damage is stronger.
