Part1Expression of neural stem cell-related antigens in SD rat brainObjective To observe and analyze expression of neural stem cell-related antigens(nestin,CD15,NG2)in SD rat brain during different developmental phase.Methods each age group(7days after birth,3months old)SD rats wereanesthetized, perfusion with fixative (4%paraformaldehyde) and brain removal, fixed4hours with paraformaldehyde, then10%,20%sucrose progressively dehydrated, and thenfrozen coronal tissue sections were obtained in accordance with the 《rat brain slice feedand map reading》 Xuzhou Medical College, Wang Pingyu. Immunofluorescent methodwas used to observe the expression of neural stem cell-related antigens (nestin,CD15,NG2) and the distinctive marker for O-2A lineage (A2B5).Results Results of immunohistochemical staining showed that, there seldom wasexpression of nestin and CD15in White and gray matter, mainly expressed around SVZand the dentate gyrus of hippocampus, positive expression was lower with the age growth.NG2has been found in different brain regions in observed all age groups, Different fromearly stage, some progenitor cells in sub-ventricular zone seldom expressed NG2, highexpression of NG2was also found in peripheral part of mature rat brain.Conclusions During differentiation, neural stem cells migrate gradually toperipheral part of rat brain, nestin lost firstly, then CD15gone, NG2lost slowest. Part2Expression of neural stem cell-related antigens in O-2A glia lineageObjective To observe and analyze expression of neural stem cell-related antigens(nestin,CD15,NG2)in O-2Aglia lineage.Methods Based on the differential properities of developmental time-course andcellular adhesions in O-2A glia lineage, O-2A lineage cells were isolated with a standardsharking method, and then purified with the differential adhesion method using theserum-free medium containing growth factors, which resulted in obtaining purified T2A.then, Neural stem cell-related antigens(nestin,CD15,NG2)were used to determine theexpression of the differentiated cells.Results Expression of nestin was high in the OPC (1d,2d), expression of T2A (1d)has been slightly reduced (only70-80%expression), there seldom was expression of nestinin the fully differentiated T2A (3d). Similarly, expression of CD15was higher than T2A inOPC. however, it is interesting that expression of CD15was decreased but little expressedin the fully differentiated T2A. High expression of NG2was found in the O-2A lineageglia (O-2A precursor cell, T2A).Conclusions During developmental progress of O-2A lineage cells in the centralnervous system, biomarkers of O-2A lineage cells were attenuately expressed from immaturestatus to mature status. Different neural stem cell marker expression pattern is different.
