Research On Molecular Mechanism Of Shuizhongcao Over Recurrent Oral Ulcers

Objective Through animal experiments observed Shuizhongcao particles treatment of ROU, determines the interleukin-4(IL-4)concentration change by ELISA to study the change of inflammation, determines the interleukin-8 (IL-8/CLCX8),granulocyte-macrophage colony-stimulating factor GM-CSF concentration change to expound the immunological mechanisms in the reversal of the reduction of netrophils.Methods①70 healthy clean level SD rats, female and male in half, weight 180±20g,6-8 weeks, were randomly divided in to 5 groups:normal group,0.9% NS treated group, Shuizhongcao particles 5 days group, Shuizhongcao particles 10 days group, Shuizhongcao particles 15 days group.Except of normal group, other groups fluorouracil injection (5-FU) abdominal cavity chemotherapy in the first and third day, abdominal injection with 10% dose of chloral hydrate by 0.3ml/100g of SD rats in the fourth day, separate the upper and lower jaws with hemostat clamp, pull out the right side of the oral mucosa with tweezers gently, clamped the NaOH crystal diameter of about 5mm with tweezers, accurate placed in the right buccal mucosa, contacted continually for 5-10 seconds fixedly, take the naked eye seeing mucous membrane broken with bleeding as the standard, established the model of oral ulcers.②Check normal and damaged oral mucosa, and do pathological examnation, observe the disease and recovery of ROU.③All groups give normal diet, observing generals symptoms and the oral ulcers change of rats in each group, executed the corresponding group rats on the 6th,11th and 16th day, detect all groups rats peripheral blood of IL-4,IL-8 and GM-CSF level.Results①Observation of oral mucosa pathological organization showed that:the normal group rats' soomth, no obvious inflammatory cell infiltration.Other group rats oral mucosal ulcers can see the mucosal epithelium burst and fall off, formed the ulcer. Surface fiber exudation,necrotic cells and inflammatory cells together formed pseudomembrane covering, inflammatory cells is mainly neutrophils and lymphocytes, followed by plasma cells.②Compared with 0.9% NS control group,Shuizhongcao particles treatment group rats' recovered well, oral ulcers healing fast 1-2 days.③To determine the content of IL-4,IL-8,GM-CSF in the Rats serum by ELISA, compared with other groups,IL-4 levels in Shuizhongcao particles 5 days group significantly increased,,P<0.05; IL-8 levels in Shuizhongcao particles 10 days group, Shuizhongcao particles 15 days group significantly reduced, P<0.05; GM-CSF levels in Shuizhongcao particles 5 days group significantly increased, P<0.05.Conclusion①Shuizhongcao particles can promote the oral ulcers healing, improve general situation of rats.②Shuizhongcao particles can promote the body secreting IL-4, inhibit the IL-8 secretion, suppress the inflammatory reaction. The GM-CSF secretion increased at the early inflammation, decreased at the later stage of the inflammation,can't show that Shuizhongcao particles promote the GM-CSF secretion.③The healing of recurrent oral ulcers after chemotherapy has important relationship with the elimination of inflammation. Shuizhongcao particles can promote the the elimination of inflammation, accelerate oral ulcers healing.
