Objective:To discuss the protective effects on hepatic ischemia reperfusion injury (IRI) and the influence on the Nrf2/Keap1-ARE signal pathway by oridonin preconditioning in rats after extracorporeal liver resection and partial liver autotransplantation.Method:Establish extracorporeal liver resection and partial liver autotransplantation model of rats. 54 male Sprague-Dawley rats were randomly divided into three groups on the basis of the different drugs administrated before liver resection: 18 rats in sham-operated group (SO),18 rats in control group with saline pretreatment (CT) and 18 rats in oridonin preconditioning group (OP). The animals were killed at 2h , 6h , 24h after autotransplantation liver reperfusion or sham operation respectively. Plasma samples were gathered for AST and ALT test; Part of the liver tissues were made in paraffin-embeded speciments for detecting histological change (HE). Liver tissues MPO,SOD,GSH and GSH-Px activities were determined. The liver Nrf2 and HO-1 expressions were defined by western blot analysis.Results: Firstly,compared with the SO group,CT group at 2h,6h,24h after partial liver autotransplantation reperfusion:(1) Liver was obvious edema,hemorrhage,and neutrophil infiltration;(2) Liver MPO activity was increased(P<0.01),SOD activity was decreased(P<0.01),Liver GSH and GSH-Px levels were significantly decreased (P<0.01,P<0.01);(3) Serum AST and ALT were significantly increased (P<0.01,P<0.01);(4) Expressions of Nrf2 and HO-1 in the 1iver tissues were enhanced (P<0.01,P<0.01). Secondly,compared with the CT group,OP group at 2h,6h,24h after partial liver autotransplantation reperfusion:(1) The pathological injuries of liver was improved;(2)Liver MPO activity was decreased(P<0.01),SOD activity was significantly enhanced (P<0.01), Liver GSH and GSH-Px levels were elevated significantly (P<0.01,P<0.01);(3) Serum AST and ALT were significantly decreased(P<0.01,P<0.01);(4) Expressions of Nrf2 and HO-1 in the 1iver tissue were increased (P<0.05,P<0.01).Conclusion: (1) Extracorporeal liver resection and partial liver autotransplantation can lead to severe hepatic ischemia reperfusion injury,the augment of oxyradical is one of the main causes. (2)Oridonin pretreatment has significantly protective effect on hepatic ischemia reperfusion injury induced by extracorporeal liver resection and partial liver autotransplantation , attributable to the protection through activation of the Nrf2/Keap1-ARE signal pathway.
