Objective To observe the effect of perfluorocarbon by intraperitoneal pre-injection on rats with myocardial ischemia in the expression of monocyte chemoattractant protein-1(MCP-1),interleukin-6 (IL-6),nuclear factor kappa B (NF-κB) and investigate the possible protective effect and mechanism of perfluorocarbon on ischemic myocardium.Methods The 50 male Wistar rats were randomly divided into 5 groups (10 rats/each group):the sham group, ischemia group,low dose group, middle dose group and high dose group. The rats in the sham group and ischemia group received perfluorocarbon 5ml/kg daily by intraperitoneal injections before model building, the rats in low dose,middle dose and high dose group respectively received perfluorocarbon 5ml/kg daily,15ml/kg daily,30ml/kg daily by intraperitoneal injections before model building; Rat heart ischemia was established by the ligation of the left anterior descending coronary artery except the sham group.The level of MCP-1,IL-6 in peripheral blood of each group was detected respectively by enzyme linked immunosorbent assay (ELISA) in different ischemic times(20min,40min,60min),myocardium were assayed by Hematoxylin and Eosin(HE) staining to observe the pathological changes after take blood last time,immunohistochemistry were carried out to investigate the activation of nuclear factor-KB.Results The level of MCP-1,IL-6 increased gradually with the ischemic time lasting, Compared with sham group, the level of MCP-1,IL-6 were significantly increased(P<0.05) in ischemia group;at 40min and 60min ischemic time point,the level of MCP-1,IL-6 in middle dose group and high dose group were significantly lower than ischemia group (P<0.05),but at 20min ischemic time point, middle dose group had no statistical significance contrasted with ischemia group(P>0.05),low dose group had no statistical significance compared with ischemia group(P>0.05); in contrast with ischemia group,middle and high dose PFC reduced the histopathological changes of myocardium and inhibited the activation of NF-κB effectly(P<0.05).Conclusions Certain dose of PFC can decrease the level of MCP-1,IL-6 can inhibite inflammatory cell infiltration and the activation of NF-κB; it also can protect ischemic myocardium to a certain degree, the protective mechanism maybe have connection with the function of PFC which can inhibite NF-κB related signal transduction pathway.
