Objective To investigate the inhibition effects of SPG on mice hepatoma by giving different doses of SPG in H22 cell, and discussed the mechanism from caspase-3 and bax protein expression in hepatoma cells, proliferation of hepatoma cells, serum levels of TNF-a, tumor weight and body weight aspects.Methods Fifty mice were inoculated with hepatoma H22 cells and were randomly divided into five groups with 10 mice every group. The high, medium and low dose groups were administered intragastrically with SPG by 100,50 and 10mg/kg·bw daily respectively. The tumor control group was given the "same amount of saline daily. The cylophosphamide group was given 20mg/kg-bw cyclophosphamide by intraperitoneal injection every other day. After ten days, all mice were killed.1. Tumor weight/body weight index:The mice were weighed, and the weight of tumor tissue were measured after killed, and calculate the tumor weight/body weight index.2. Level of cell proliferation:To assay the proliferation level of hepatoma cells in mice by using MTT.3. Level of apoptosis:The immunohistochemistry was used to detect Caspase-3 and Bax protein expression.4. Serum levels of TNF-a:The serum tumor necrosis factor (TNF)-a wasmeasured by enzyme linked immunosorbent assay (ELISA).ResultsTumor weight/body mass index:The weight ratio of tumor and body of the high dose SPG group was 1.06%, and compared with tumor control group decreased significantly (P<0.05)Level of cell proliferation:The proliferation level of the tumor control group was 1.135±0.032 which was significantly higher than that of the high-dose SPG group (P<0.05)Level of apoptosis:The expression level of Caspase-3 and Bax protein in high-dose group were 40.20% and 38.10% respectively, and those of the tumor control group were 5.00% and 4.68%. There were significant differences between the groups (P<0.05)Serum levels of TNF-a:The serum concentration of TNF-a of the high-dose SPG group was 129.367±4.443 (pg/ml) which was significantly higher than that of the tumor control group (p<0.05)ConclusionSPG can inhibit the proliferation of hepatoma cells; promote the expression of caspase-3 and bax protein to induce apoptosis of hepatoma cells. SPG can inhibit hepatoma cells significantly.
