Metaplexis Japonica is a plant from Metaplexis, Perennial herb wrap, which grows at hillside, field and roadside.It's widely distributed from Hebei Province to Fujian Province. The stem and skin are used to make staple rayon. Its roots can cure traumatic injury and snake bite wound. Its leaves can cure children's tummy bug. Its fructification can cure internal lesion caused by overexertion. The nap of seed can stop bleeding. It is significant of the research on its chemical composition and biological activity.Modern extraction and separation technology were used to analyze Metaplexis Japonica's vine. With qualitative test, Metaplexis Japonica was composed of carbohydrates, protein, sterid, alkaloids, flavornoids, triterpene and cardiac glycoside. It probably have volatile oil, lactone and tannins. With tetrahydropalmatine as reference substance, total alkaloids were determined by acid-based titration. The content of total alkaloids was 0.28%. Through a test of three factors and three levels, extraction process of flavonoids was confirmed which was 90% ethanol,20 times volume of material, extraction time of 2 h. With lutin as reference substance in the extraction process, The content of total flavonoids is 1.2%. Sulfonic mercuric acetate reagent method is confirmed:RSD of precision is 0.6%, and recovery rate is 99.1%, RSD of precision is 0.9%. With pregnenolone as reference substance, sulfonic mercuric acetate reagent method was used to determine the content of C21 steroidal glycosides in metaplexis japonica and the content of C21 steroidal glycosides is 2.0%.Metaplexis japonica was extracted by 95% ethanol. With Silica gel column and sephadex LH-20 column, compound L-1 and compound L-2 were abtained. Combining With IR, NMR, MS to identify the structure, the molecular weight of L-1 is 468.4.
