Cohesin, a hetero-tripartite complex of SMC1, SMC3 and Rad21, associates with chromatin after mitosis and holds sister chromatids together following DNA replication. It has been reported that SMC1A, a subunit of Cohesin, was mutated in human colorectal cancer, which induced genome instability and cancer.However, so far there hasn't been any reports investigating the role in glioma. In this study, we found that SMCIA is highly expressed in human glioma tissues and C6 cell lines, compared with normal brain tissues and rat glial cells. In the following steps, lentiviral-mediated SMC1A shRNA were used for study. Results showed that knockdown of SMC1A in U251 cell lines by Lentiviral infection induced cell growth inhibition and G2/M phase cell cycle arrest, with increased CCNB1IP1 mRNA levels in U251 cells, decreased CCNB1 and PCNA expression. These results suggest that Kowndown of SMC1A expression inhibit U251 growth. Therefore, SMC1A could be a potential target for its therapeutic application in glioma.In the article of TC10β/CDC42 GTPase activating protein is required for the growth of cortical neuron dendrites, All constructs are proved to be effective both in 293T cell and in neurons.
