Objective To study the relationship of action of matrine on mouse fibroblasts L929 cells and eIF4E,caspase-3 mechanisms.Methods The L929 cells were treated by different concentration of matrine. the concentration were as follow:0.25g/L,0.50g/L.0.75g/L,1.00 g/L. The viability of L929 cells were measured by MTT assay. Expression of phospho-eIF4E, phospho-eIF4E-BP 1 and caspase-3 were determined by Western blot analysis.Results The factorial design analysis of variance results:FC*T=1.027, P=0.417, the time and concentration have no interaction with each other. At the same time comparison between the different concentrations and different times of the same concentration were compared between the single factor analysis of variance. The role of different concentrations of matrine 24,48,72h, respectively, on the growth of mouse fibroblast L929cells,at the same time, following the increasing of concentration of matrine, the survival ratio to L929 was reducing (F=31.41,34.15,41.55, p<0.01); at the same concentration, following the lasting of time, the activity of L929 was not significantly decreased. Matrine could inhibit the proliferation of mouse fibroblast L929 cells, decrease the expression of phospho-eIF4E and phospho-eIF4E-BP1 and increase the expression of caspase-3.Conclusions Matrine could induce the apoptosis of L929 cells, its role is to eIF4E,eIF4E-BP1and caspase-3 pathway.
