| Radix Aconiti comes from of the root of Aconitum pendulum Busch and A. flavum Hand.-Mazz. of which family Ranunculaceae Aconitum plants, the modern study found that Radix Aconiti had effect on antipyretic, antiphlogistic, analgesic, local anesthesia and bacteriostasis. A. flavum Hand.-Mazz.. As one medicinal plant of the Radix Aconiti, it was widely used treatment injuries pain, rheumatism arthritis pain, trauma, external bleeding and other symptoms, in its good treatment function at the same time, the cardiac toxicity limited the safe use and effective use, in order to improve the curative effect, reduce the toxicity, and for the safety of the effective and clinical applications for preparation, we had been a preliminary research on the processing methods.This topic mainly researched two methods to lower the toxicity of the A. flavum Hand.-Mazz., First of all, this thesis summarized the research results in recent years. Secondly, identified 2 different process methods on A. flavum Hand.-Mazz. and get 8 different processed products according to the methods of the family Ranunculaceae Aconitum and principles of processing methods science of processing Chinese materia medica, acute toxicity experimental research were made on crude drug and processed products, the median lethal dose of crude drug was 1.202g/kg, the median lethal dose the process of oven-dry 105℃after 5 hours was 4.855g/kg, the median lethal dose the process of oven-dry 105℃after 7 hours was 5.770g/kg, steamed products of 3days soak showed obvious side effects the maximum volume of 320g/kg, other prepared products were no obvious abnormalities. Fanally, optimazed the extraction process of the total alkaloids, using the acid dye colorimetry method in analyze and the result is 0.521% of the crude drug, we established a method that is simultaneous determination for contens of aconitine and 3-acetylaconitine from the crud drug of A. flavum Hand.-Mazz. by HPLC, The results showed that the content deteminations of aconitine and 3-acetylaconitine are separately 2.115mg/g 0.123mg/g, after different preparations, the content of total alkaloids,aconitine and 3-acetylaconitine reduced on different degrees of 8 processed products。By using acute toxicity experimental research and determine the content of the main chemical element, we prepared preliminary study on the processing methods of A. flavum Hand.-Mazz., demonstrated all the process methods we had done can lower the toxicity, Increased the safety use value,provide a basis theoretical foundations for the reasonable processing technology standards and clinical practice. |
