Objective To research the influence of the speed of rising of blood glucose concentration in rats with brain damage caused by hypoglycemia induced by insulin. Put forward the"brain injury induced by glucose-reperfusion",to provide some suggestion for the treatment of hypoglycemia.Methods 30 adult male Wistar Rats were randomly assigned to experimental group,medicinal control group I,medicinal control group II,blank control group.Experimental group were intraperitoneal injected with insulin. Hypoglycemic rats were induced by insulin.The blood sugar level is lower than 1mM and sustaining 1 hour. According to the blood glucose concentration after glucose reperfusion ,the experimental group was divided into 1~2mM,5~10mM,10~15mM group and every group has five rats.The medicinal control group I were prepared by intravenous administration of 1.5 ml/h of 25% glucose simultaneously with 15U/kg of insulin to maintain a plasma glucose level between 5 and 10 mM for 5 hours. The medicinal control group II were prepared by intravenous administration of 1.5 ml/h of 25% glucose to maintain a plasma glucose level between 10 and 15 mM for 5 hours. The blank control group were normal . HE,TUNEL staining were used to detect the neurons of the hippocampus in hypoglycemia and glucose-reperfusion(HG/GR) rats.Results Brain damage were detected in all rats. (1) HE stain ,Compared with the control group, the number of the normal neurons are more in the experiment group. There was statistical significance between control group and experiment group(P<0.05). . The normal cell percentage in medicinal control group I,medicinal control group II,blank control group is 94.8%,95.2%,94.7%,there wasn't statistical significance(P>0.05). Compared with the 1-2mM group and 5-10mM group, there are fewer normal neurons in the 10-15mM group(p <0.05). (2)TUNEL stain,compared with the control group ,the apoptotic index are higher in the experiment groups.There has significant difference between control group and experiment group(P<0.05). The apoptotic index in medicinal control group I,medicinal control group II,blank control group is 2.1%,2.4%,1.3%,there wasn't statistical significance(P>0.05).Compared with 1-2mM group and 5-10mM group, the apoptotic index are higher in the 10-15 group(P<0.05).Conclusions Our results suggest that at the same level of blood glucose and the same time of the hypoglycemia ,the speed of rising of blood glucose concentration in rats is related to the brain damage caused by hypoglycemia . When the speed of rising blood glucose concentration was too quick, The hypoglycemic brain damage was aggravated. All these suggest that"brain injury induced by glucose-reperfusion"may exist.
