Objective to observe the changes of the contents of Caspase-3 and the expressions of the Caspase-3's genes with the different hypoxia times and theδ?opioid receptor agonist on the cerebral tissue of the left cortex of the rats.Methods fifty-four SD rats were randomly divided into three groups: the hypoxia one - day group, the hypoxia five-day group and the hypoxia ten-day group. Each group was divided into three small groups: the control, the hypoxia and the hypoxia-drug (UFP-512). After cutting the head of the rats and getting the tissue of the rats'left cortex, we can treasure the expressions of the Caspase-3's genes and the contents of the Caspase-3 with the RT-PCR and the western blotting technologies and compare the differences between the inter-group with the use of the statistical analysis.Results with the time prolonging of the hypoxia time, the expressions of the Caspase-3's genes increase and the contents of Caspase-3 of the tissues also increase and comparison between the inter groups ,the difference has statistic significant (p<0.05); When the hypoxia time is one - day, the expressions of the Caspase-3's genes and the corresponding contents of the Caspase-3 have no obvious change and the difference has no statistic significant(p>0.05); When the hypoxia time is five-day, the expressions of the Caspase-3's genes and the corresponding contents of the caspase-3 between the hypoxia and the hypoxia-drug group have statistic significant with the comparison of the control (p<0.05) and the Caspase-3's genes and the corresponding contents of the caspase-3 of the hypoxia-drug group is lower than the hypoxia group's and the different has statistic significant (P<0.05).Meanwhile When the hypoxia time is ten-day, the expressions of the Caspase-3's genes and the corresponding contents of the caspase-3 between the hypoxia and the hypoxia-drug group have statistic significant with the comparison of the control (p <0.05) and the Caspase-3's genes and the corresponding contents of the caspase-3 of the hypoxia-drug group is lower than the hypoxia group's and the different is obvious and has statistic significant (P<0.05).Conclusions Caspase-3 can be as a point of the damage extent of the chronic hypoxia to the brain neuron and as a new type ofδ-opioid receptor agonist, UFP-512 can activiateδ-opioid receptor and reduce the exprrssion of the Caspase-3 and play a part of the brain-protecting.
