Objective: We used immunohistochemistry to investigate the expression of YAP and Survivin in normal endometrium,atypical hyperplasia and endometrioid carcinoma, to analyse the relevance of the two and its relationship with ER, which can provide the clinical reference for the endometrial carcinoma's early diagnosis and appraisal prognosis.Methods: Surgically resected specimens of 45 cases of endometrioid carcinoma, 20 cases of atypical hyperplasia, 20case of normal endometrium were collected from No.1 Hospital of Dalian Medical University. All of the cases have no radiotherapy and chemotherapy before surgery. All of the endometrioid carcinoma samples have the ER examination expression results in our pathology department. Statistical method: Uses SSPS16.0 the software package to carry on the analysis. We use the x2 examination, Fisher test and Spearman to analyze the experimental result.α=0.05 is the examination standard, P < 0.05 has statistical significance.Results:(1) The positive rates of YAP and Survivin in atypical hyperplasia and endometroid carcinoma are significantly higher than that in normal endometrium, P <0.05; There was no statistical difference between atypical hyperplasia and endometroid carcinoma, P>0.05. The positive expression of YAP were statistically associated with surgical-pathological stage (P <0.05). There were no statistic difference expression in patients'age, histological grade ,myometrial invasion and lymph node metastasis, (P >0.05). The positive expression of Survivin statistically associated with surgical-pathological stage, histological grade, lymph node metastasis, (P<0.05). There were no statistic difference expression in patients'age and myometrial invasion (P >0.05). (2) In 45 cases of endometroid carcinoma, The positive expression of ER statistically associated with histological grade, and myometrial invasion (P <0.05). The expression of YAP and Survivin is positively correlated, r=0.573, P <0.05.The expression of YAP and Survivin with ER are negative correlated, P <0.05.Conclusions:1,The positive rates of YAP in atypical hyperplasia and endometriod carcinoma were significantly higher than that in normal endometrium, suggesting that the overexpression of YAP might make the Hippo pathway out of control, break the balance of cell proliferation and apoptosis, which could cause the endometriod carcinoma's development.2,The positive rates of Survivin in atypical hyperplasia and endometriod carcinoma were significantly higher than that in normal endometrium; The positive expression of Survivin statistically associated with surgical-pathological stage, histological grade, lymph node metastasis, suggesting that the overexpression of Survivin might reflect the endometriod carcinoma's malignant degree.3,The positive expression of ER was statistically associated with histological grade, and myometrial invasion, suggesting that ER might be one of the marks which could reflect the endometriod carcinoma's prognosis.4,The expression of YAP and Survivin in endometrioid carcinoma was correlated, YAP might induce the expression of Survivin, promoting the cells proliferation and anti-apoptosis. YAP and Survivin might participate in endometrioid carcinoma's development. The higher of YAP and Survivin expressed, the lower of ER expressed, suggesting that detect the three marks together might help the early diagnosis and appraisal the prognosis of endometrial carcinoma. |
