Objective:To compare and analyze the protein expression profiles of extensively drug-resistant (XDR) and multidrug-resistant(MDR)tuberculosis (TB) isolates, and identify differentially expressed protein spots related to XDR-TB, and to detect related genes expression changes in Mycobacterium tuberculosis XDR/MDR and the standard strain H37Rv.Methods:Total proteins extracted from clinical isolates of XDR-TB and MDR-TB as well as the standard mycobacterium tuberculosis H37Rv were analyzed with two-dimensional electrophoresis (2-DE). The Progenesis SameSpots program was used for analyzing the 2-DE images to identify differentially expressed protein spots in XDR-TB compared to MDR-TB and the standard strain. Some selected protein spots were used for the MALDI-MS peptide mass fingerprinting.Results:The results of this study showed that protein expression profile of XDR-TB was significantly different from those of MDR-TB and the H37Rv standard strain. There were 53 differentially expressed protein spots in XDR-TB. Fifteen proteins differentially expressed in XDR-TB were identified by MALDI-MS peptide mass fingerprinting. The changes of some protein spots were detected in XDR, MDR and H37Rv strains by using real-time PCR, and The "210 strains of Mycobacterium tuberculosis nucleic acid hydrolase iunH " was confirmed in MDR strains.Conclusion:Protein expression profile of XDR-TB is different from the one of MDR-TB and the H37Rv standard strain, and the differentially expressed protein spots could be used as potential gene targets for further investigations to study their functions in XDR/MDR-TB.
