Differential Expression Of Plasminogen Activator Inhibitor-1 In Hypertrophic Scar Versus Normal Skin

Objective:To investigate the mechanism of Plasminogen activator inhibitor-1(PAI-1) on hypertrophic scar by comparing PAI-1 expression between hypertrophic scars and normal skins.Methods:The human hypertrophic scars and normal skins were taken from patients during operation. Samples were trimmed by sharp dissection to remove adipose tissue and epiderm.Total RNA from hypertrophic scars and nomal skins were extracted.The quality of RNA was analyzed in 1% agarise gels stained with ethidium bromide.and the OD values was determined by spectrophotometric analysis at 260 and 280nm. The expression of PAI-1 in hypertrophic scar and normal skin was analyzed with real-time PCR.Results:The quality of RNA was valid.The solubility curve of PAI-1 andβ-actin was unimodality. The expression of PAI-1mRNA was about 2.7 fold in hypertrophic scar versus normal skin.lt was elevated significantly in hypertrophic scar (P<0.05)Conclusion:PAI-1 is up-regulated in hypertrophic scar,which may play a important role in the development of hypertrophic scar.
