This paper investigated the antioxidation of the extract from Eucommia Ulmoide Oliver Leaves in vitro and in vivo, and the technique of microencapsulation of the extract from Eucommia Ulmoide Oliver Leaves was also further studied. The results were as following:1. The total content of flavonoids extracted with water was 32.27%.The antioxidation of Eucommia Ulmoide Oliver Leaves extract was tested in vitro. The result indicated that Eucommia Ulmoide Oliver Leaves extract had the high antioxidative activity. There was linear correlation between the TAC (Total antioxidant capacity) and total flavonoids, the calculated equations was y=37.924x+0.7928. The extract had good ability to eliminate superoxide anion(O2·-), hydroxyl radical (·OH), DPPH free radical, and the calculated IC50 was 0.474,414,0.357mg/mL reseparately. The extract had also good inhibitory ability on lipid peroxidation and DNA damage, and the calculated IC50 was 0.413,0.124mg/mL reseparately. The TAC of the extract from E U Oliver Leaves was higher than that from Ginkgo Biloba, but less than that from green-tea and grape seed.2. The influence of the extract from Eucommia Ulmoide Oliver Leaves on rats was observed during four week feeding period. The results showed the rats body weight increased steadily with the increasing doage of E U Oliver extract, and this was also observed for the weight ratio of heart to body with significant difference between dosages, but did nor find in liver, kidney, spleen ratio.The antioxidant ability indexes of serum and liver showed the same trend with feeding the extract of E U Oliver, and showed dosage-effect dependent relationship with the most significant for TAC, although there were significantly effects on MDA reduction and SOD activity enhancement of serum and liver, and almost no effect on GSH—Px, CAT activities.3. The acacia and (3-cyclodextrin on encapsulation for the extract of E U Oliver was investigated and the factors involved in microencapsulation were also tested. The both single factor and orthogonal test indicated that acacia andβ-cyclodextrin could embedded extraction well, and the influencing ability from great to small was in the following order: core material/wall material ratio>feed rate>wall materials'ratio>extract oncentration. The summarized optimal parameters for the microencapsulation of the extract were 2:1 for the ratio of acacia/β-cyclodextrin,0.5:1 for core material to wall material,15% material concentration with feeding rate 20ml/min.
