The dry root tuber of Ophiopogon japonicus (L.f.) Ker-Gawl.(Liliaceae) is one of the most important Triditional Chinese Medicine. Elaeocarpus sphaericus (Gaertn.) K. Schum. (Elaeocarpaceae) is a tall tree growing in tropical areas of China.The 95%ethanol extract of O. japonicus and E. sphaericus were studied respectively. On the basis of modern chromatography isolation techniques and spectroscopic structure identification methods, thirteen homoisoflavonoids were isolated with their structures identified from O. japonicus, including 8-Formyl-7-hydroxy-5,4'-dimethoxy-6-methylhomoisoflavone (1). 6-Aldehydoisoophiopogonone B (2), Ophiopogonone C(3), Methylophiopogonone B Monomethyl Ether(4),8-Formylophiopogonone B(5), Ophiopogonanone B(6), 5,7,4'-Trihydroxy-3'-methoxy-6,8-dimethylhomoisoflavanone(7), Methylophiopogonanone B(8), Methylophiopogonanone A(9), 8-Formyl-7-hydroxy-5,4'-dimethoxy-6-methylhomoisoflavanone (10), 8-Formylophiopogonanone B(11), Methylophiopogonanone B Monomethyl Ether (12),8-Formylophiopogonanone A(13). Among these homoisoflavonoids, compounds 1,2 and 11 are new compounds. The NMR data of compounds 10 was first reported here and compounds 5 in previous literature were reassigned definitely by 2D NMR experiments.Meanwhile,the cytoprotection activities of the above 13 compounds were assayed based on H9C2 cell which was is chemic and hypoxic.Five indolizidine alkaloids were isolated from E. sphaericus. Their structures were determined as (±)-3-oxo-isoelaeocarpine(14), (±)-elaeocarpine-N-oxide (15), (±)-isoelaeocarpine (16), (±)-elaeocarpine (17) and (-)-isoelaeocarpiline (18) by physicochemical propertiesand spectral data. Compounds 14 and 15 are new compounds. Enantiomers of compounds 14 and 16 were separated on a chiral CD-Ph column and determined by TDDFT quantum chemical calculations of their ECD spectra. In conclusion, the current study has provided rational and experimental basis for further development of these plants and their quality control.
