Abstract/Summary:
Objective: Anti-tumor function mechanism will be discussion of Gingko leaf extraction Gingko leaf total flavanone EGb761. The influence of bladder cancer cell T24 come from THP union gingko total flavanone EGb761, as well as the effect of T24 cell acting on caspase survivin gene expression. Thus to provide the theory basis for the clinical application of xingding as bladder perfusion adjuvant chemotherapy drugs in bladder perfusion chemotherapy. Examines the expression situation of Survivin and the caspase-3 gene in T24 cell after use the RT-PCR law, various densities medicine affects 48h. Methods : Bladder cancer T24 cell be raised in the incubators in carcinoma 37 oC, bladder cell line T24 be trated with different density gingko leaf total flavanone compound injection(Xingding ? Yinxingdamo injection) and unifies THP ( bladder cancer clinical irrigation chemotherapy medicine); with the MTS method to describe cell growth curve so that clear the function about the gingko leaf total flavanone in bladder cancer chemotherapy , with RT-PCR method to detect the expression situation of the Survivin and caspase-3 gene in T24 cells after it was effected 48 h by various densities medicine.Results:1. It was showed that the cell T24 concentration and time respectively on bladder cancer cell proliferation inhibition is more obvious when intervent T24 by different concentration of XING Ding and THP. And concentration dependent effect relationship is obvious (P<0.05), and it is obvious that inhibition of T24 cell proliferation with XING Ding combined THP (P<0.05).2. The results of RT-PCR detection of Survivin mRNA showed that: the Survivin mRNA expression be cut of after intervention T24 cell by XING Ding combined with THP (P<0.05), than a separate application of xingding or separate application of pirarubicin (P<0.05).3. The results of RT-PCR detection of Caspas-3 mRNA showed that: the Caspas-3 mRNA expression increased after intervention T24 cell by XING Ding combined with THP (P<0.05), and it is obvious to use XING DING combined with THP(P<0.05). Conclusions: Xingding combine with THP can enhance the function of inhibition of T24 cell proliferation, and the result is better than THP. Its molecular mechanism is possible related with the suppression of the Survivin expression and the increasing of the Caspas-3 gene expression, yet to be confirmed by further studies.
