Age Effect On Wnt-1 And OPG Expression In Periodontal Ligament Cells Of Pressure Side

Objective: Through the establishment of rats orthodontic tooth movement model, using immunohistochemistry and RT-PCR to detect OPG and Wnt-1's expression in rat periodontal ligament (PDL)cells. Explore age factor's molecular mechanism in orthodontic bone rebuilding. Methods : Adopt Ni - Ti traction spring establish rats to the first molar maxillary right in the middle of the moving orthodontic reason move models, Every day observe rats diet and health, detection of orthodontic device ,Preparation of orthodontic tooth movement model of rats, 80 wistar experiment rats were divided into two group, youth group 40, junior 40 , each group take 20 only used to organize morphology research, 20 only for RT - PCR determination. The hematoxylin-eosin stain observation rats periodontal tissue morphological changes,The experession of OPG and Wnt-1 was detected by immunohistochemistry and RT-PCR technique at 1,3,7,14d. Results:①On day 3,pressure side of periodontal membrane lumen narrowed,Near the alveolar periodontal membrane appear osteoclast;On day 7,pressure side of periodontal membrane lumen further narrowed,After orthodontic periodontal tissue form change,Display model success.②Young rats in the teeth move 1, 3, 7, 14d, OPG are expression on the pressure side,but on day 3 stronger than 1,14d;On day 3 and 7d OPG expression positive cells on the pressure side of the young rats were much stronger than that of aged ones. Immunohistochemical results and PCR results are basically the same.③Both youth and adult groups rats, Wnt-1 were no expression in PCR,but on day 3 Wnt-1 expression positive cells of the young rats. Conclusions: In orthodontic force under the action of the lateral pressure, OPG close relations with age expression, OPG expression increased in press area in a time - dependent manner; this provides a good point for orthodontic treatment on teeth movement speed and alveolar bone rebuilt aspects of the control ;Age factor is an important factor in bone rebuilding; Wnt-1with orthodontic bone rebuilt correlation need to be further researched, We Can detect other osteogenesis relevant factors such as: Wnt - 3a, Wnt - 4, Wnt - 5a, to further testing Wnt and orthodontic bone rebuilt correlation.
