Influence Of Salvia Miltiorrhiza Beg.f.Alba Pharmacological Preconditioning On BCL-2 And Bax Of Middle Cerebral Artery Obstruction

Objective:To investigate the effects of pharmacological preconditioning with different does of aqueous extract of salvia miltiorrhiza beg.f.alba on apoptosis and expression of Bcl-2 and Bax of neurons following middle cerebral artery obstruction. To explore protective effects of salvia miltiorrhiza beg.f.alba pharmacological preconditioning on the neurons and the optimal dosage.Method:Before the Middle cerebral artery obstruction(MCAO) models, rats were randomly devided into four groups: one contrast group(n=9)and three therapy groups with different dose of salvia miltiorrhiza beg.f.alba(low dose,middle dose,and high dose) for 7 days. Then the rats were killed 24 hours after MCAO and expression of apoptosis related protein Bcl-2 and Bax of the brain slice was detected by immunofluorescence staining.Results:Expression of apoptosis related protein Bcl-2 and Bax:Bcl-2 positive cells were mainly stained with green fluorescence in cytoplasm,nuclear membrane and apophysis. The expressions of Bcl-2 and Bax in the sham group were low.Compared with the contrast group, the expression of Bcl-2,the strength of green fluorescence was higher in high dose plasma concentration of salvia miltiorrhiza beg.f.alba group,middle dose plasma concentration of salvia miltiorrhiza beg.f.alba group and low dose plasma concentration of salvia miltiorrhiza beg.f.alba group, the differences were significant(P<0.05). On the other hand, the Bax positive cells were mainly stained with green fluorescence in cytoplasm,nuclear membrane and apophysis. Compared with the contrast group, the expression of Bax and the strength of green fluorescence are lower in high dose plasma concentration of salvia miltiorrhiza beg.f.alba group,middle dose plasma concentration of salvia miltiorrhiza beg.f.alba group and low dose plasma concentration of salvia miltiorrhiza beg.f.alba group, the differences were significant(P<0.05).Conclusion:1.Preconditioning by different aqueous extract concentrations of salvia miltiorrhiza beg.f.alba could significantly decrease apoptosis cells.2. The mechanism may be involved increase of Bcl-2 expression, decrease of Bax.
