ObjectiveTo further improve the physico- chemical and chemical properties through the study on peripheral nerve coloboma with the polylactides catheter with different permeabilities, enhance the clinical application of peripheral nerve coloboma .MethodThe polylactide are preparied through the Open-loop polymerization method of Lactide and pouringed translucence catheter through controling the sodium chloride'size in sterile conditions. Polylactide-aperture'size are 0,0~10um,10~100um, length: 10mm ,thickness:0.2~0.4mm, inside diameter:1.5mm.Health wistar rats 50,male,weight between 200~300g,randomly divided into 5 groups,each with 10. The rats are all anesthetized by ketamine as the method of intraperitoneal injection and revealled left leg sciatic nerve stem in sterile conditions.The length of nerve ends is 8mm as their natural retract after respectively cutting 6mm nerve of small pieces.Five group for the experiment: Group A:autologous nerve transplantation, Group B1:Catheter-wall'size is 0 um, Group B2: Catheter-wall'size is 0~10 um, Group B3: Catheter-wall'size is 10~100um, Group C:The catheter is preparied by silicon.Sciatic -ends's adventitia and catheter-wall are sutured with untrauma suture of 9/0 under the microscope in the Group B1,B2,B3,C , the pieces of nerve in the Group A are overturned and suture the nerve ends .We suture the wounded skin after disinfection and breed in different cage ,All rats are still live after six weeks.ResultsAll the 50 enrolled rats were analysised after sixteen weeks. (1)General observation:All experimental limb swell after one week . After sixteen weeks the basic functions of experimental crus is like the opposite side in Group A and Group B2 , the atrophic triceps of left leg improve obviously in Group B1 and Group B3. The claudication of experimental limb is still exist untill the sixteen week. Anatomical observation: After sixteen weeks, neural anastomotic smooth, nerve stem is continuous and benign, which is the diameter like the normal nerve in Group A and Group B2. The anastomotic of nerve-ends smooth in Group B1 and Group B3, and we find a lot of small blood vessels around the nerve stem,which is slightly thin than the nerve stem in Group A and Group B2. Silicon with numberous fibrous tissue are not degradation and expand in Group C.The nerve stem is minimum and thinnest in five group.⑵Nerve-electrophysiology:Including index:latentperiod,amplitude,conduction velocity(MNCV). There were difference statistically between Group A and Group B2,as the same between Group B1 and Group B3,the results of Group C are the worst in all groups,meanwhile Group A and Group B2 aret the best.⑶The recovery rate of SFI: The volume and weight are close to the normal triceps in Group A and Group B2, and are least and lightest in Group C .There were difference statistically between between Group B1 and Group B3.⑷The result of tissue and morphology: After sixteen weeks polylactide degrade completely and silicon don't degrade at all.The nerve is similar to normal nerve in Group A.The myelin and nerve fibers are regular and more than regenerative nerve in Group B1 and Group B3.There are many macrophages under microscope in Group C.⑸The count of regenerative nerve: There were difference statistically between Group A and Group B2. The count of regenerative nerve are least in Group C.Conclusion1. The catheter with different permeability is important to regenerative nerve, Semipermeable catheter make for growth of nerve and transmission of nerve fibers and recovery of limb function.2. The improvement of nerve catheter is an important significance to repair of peripheral nerve ,especially physicochemical and biological.
