Alternarialer alternata is a common and serious tobacco diseases. So far a satisfactory disease-resistant cultivar syst-em has not been established, effective prevention'drugs haven't been developed either.Currently people have been using high residual chemicals to control it,which could pollute the envir-onment and do harm to human body. On the contrary,botanical·pes-ticides are not only safe to humans, animals and environment, but also unlikely make insects and bacteria cause drug-resista-nt. They.can be easily degraded in nature,moreover they comform to agricultural sustainable development and meet the requireme-nts of people who want to improve their health. Therefore,resea-rching and developing botanical pesticides has become the main direction of new pesticides research. The purpose of this study is extracting active ingredients of antimicrobial from Radix Phytolaccae, Magnol ia officinalis and Xanthium sibiricum, develop ing Lupo compound microemulsion,establishing its quality stand-and and finishing preliminary'pharmacodynamics study by modern research technology. In order to.develop a efficient-new botani-cal pesticide which can prevent tobacco brown spot and meet the concept of "Ecological Tobacco" and the idea of environmental protection,healthy,sustainable development.During the course of extraction,taking the extraction ratio of total saponins from Radix Phytolaccae as an index, the best extraction technology has been selected. On the basis, we use macroporous resin to purify the crude extracts. The technologic-al parameters of the type of macroporous resin,adsorption cond-itions, the volumn of sample and washing,ethanol concentration, flow rate and volumn of'elution solution are taken into observe and study, and the best purifying technology has been selected. The purity of total saponins from Radix Phytolaccae can reach above 60% by using the purifying technology.It is stable, simple and suitable for industrial production. Taking the extraction ratio of Magnolol'and Honokiol as indexs,the best extraction technology of Magnolia officinallis has been selected by HPLC. Taking the extraction ratio of Chlorogenic acid as an index,the best extraction technology of of Xanthium sibiricum has been selected by HPLC. These extracting and purifying processes are high extraction yield, good stability, convenient operation and suitable for industal production.During the course of manufacturing technology study,mix pr-oportion of the extracts from Radix Phytolaccae, Magnolia offic-inalis and Xanthium sibiricum, types and amount of emulsifiers and cosolvent,prepraration technology are taken into research. Furthermore, water quality, emulsion stability, stability over time, stability of heat storage, low temperature stability, the scope of pH value of the Lupo compound microemulsion are inves-tigated. Then the formulation,manufacturing technique and the technical indicators of the Lupo compound microemulsion'are co-nfirmed. The results show that the preparation method is,simple, stable,convenient to use.During the course of quality standards study,multi-index are first used to control the quality of Lupo compound microem-ulsion. Radix Phytolaccae, Magnolia officinalis, Xanthium sibiric-um in Lupo compound microemulsion are identified by TLC, then we established the TLC identification method. The method is simple, rapid, reproducible and the spots are clearly visible,negative non-interference. The HPLC method is used to determine the cont-ent of the active ingredient of Lupo compound microemulsion. The method of determination of magnolol,honokiol, chlorogenic acid is established by HPLC.This method is accurate,reproducible and can be used to control the quality of the preparation.In preliminary pharmacodynamics study,filter paper method is used to determine the activity of spore germination of Toba-cco Brown Spot bacteria and mycelial growth rate method is used to determine the activity of Tobacco Brown Spot bacteria.It is the first time that we completed the preliminary pharmacodynam-ics study of Lupo compound microemulsion. The results show that Lupo compound microemulsion can observably inhibit spore germi-nation and mycelial growth of Tobacco Brown Spot bacteria. The higher the concentration,the more significantly reduction of the sporulation and the more obviously inhibition of the mycel-ial growth. The research has laid the foundation for further indoor pharmacodynamic experiment and field efficacy trials of Lupo compound microemulsion.
