Objective: To investigate GSK-3βandβ-catenin expression and its significance in the tissues of Diffuse large B cell lymphoma as analyzed by immunohistochemistry.Methods: Sixty three paraffin-embedded Diffuse large B cell lymphoma tissue samples were selected from pathology files of the Taixing People's Hospital,from september,2000 to september, 2010. And the diagnostic criteria were based on those described in the World Health Organization Classification Scheme 2000. There were 50 men and 13 women, and the median age at diagnosis was 54.4 years (range from 13 to 86 years). All patients were untreated before,with clear pathological diagnosis, and have been determined the sites of tumor invasion by medical, surgical, or imaging examination . We adopted Ann Arbor staging system. All patients were treated with more than two courses of CHOP chemotherapy,with or without radiotherapy. We also collected 14 cases of reactive hyperplasia tissues of lymph node as controls.We detected pGSK-3βandβ-catenin protein in Diffuse large B cell lymphoma tissue by immunohistochemistry(SP ) .Results:1.β-catenin main located in cytoplasm and nuclear(figure1,2),positive expression ofβ-catenin in DLBCL was 28.57%(18/ 63),and zero in reactive hyperplasia tissues of lymph node . There was dominant significance between them(Correctionχ~2 =5.220,P<0.05).Nuclear expression ofβ-catenin in DLBCL was14.29%(9/63),but none in reactive hyperplasia tissues of lymph node .2.pGSK-3βlocated in cytoplasm and nuclear(figure3,4,5).Positive e xpression of pGSK-3βin DLBCL was 38.10%(24/63),7.14%(1/14)in reactive hyperplasia tissues of lymph node.There was dominant significance between them(Correctionχ~2 =5.220,P<0.05).3.Correlation between pGSK-3β(Ser-9) andβ-catenin positive expression in DLBCLz In the Sixty three cases,eighteen cases both expreessed pGSK-3β(Ser-9)andβ-cateninprotein.Six cases only expreessed pGSK-3β.Expression of pGSK-3β(Ser-9)andβ-catenin in DLBCL was significantly positively correlated(r=0.767, P<0.01)Conclusions:During the development of Diffuse large B cell lymphoma,GSK-3βandβ-catenin played an important role. The possible mechanism was that GSK-3βactivity decreased after serine 9 residue phosphorylation.It preventedβ-catenin phosphorylation. Cytoplasmicβ-catenin increased, and moreβ-catenin translocated into the nucleus, activating a series of target gene transcription of cell proliferation, differentiation ,and so on , resulting in Diffuse large B cell lymphoma.The research laid a solid foundation for our further research of GSK-3βandβ-catenin in Diffuse large B cell lymphoma from the protein, nucleic acids and gene level .It also provides a new way for the future treatment of Diffuse large B Cell lymphoma .
