Objective: To investigate the expression of E-Cadherin, CXCR4 and SDF-1 in gastric cancer and its relationship to clinicopathological characteristics by tissue microarray.Methods: We detect the expression of E-Cadherin, SDF-1 and CXCR4 in 175 cases of gastric cancer and 40 cases of non-tumorous gastric tissue by tissue microarray and immunohistochemistry.Results: (1) The positive expression ratios of E-Cadherin in non-tumorous gastric tissue and in gastric cancer were 57.5% and 40% respectively have significant difference. The E-Cadherin expression level negatively correlated with the metastasis of lymph nodes and clinical stage while no significant difference was observed in depth of invasion and the size of tumor.(2) The posive expression ratios of CXCR4 in non-tumorous gastric tissue and in gastric cancer were 25.0% and 49.7% , respectively having significant difference(P<0.05). The CXCR4 expression level positively correlated with metastasis of lymph nodes and clinical stage respectively while no significant difference was observed in depth of invasion, grade of differentiation and the size of tumor.(3) The positive expression ratios of SDF-1 in non-tumorous gastric tissue and in gastric cancer were 40% and 57.7%, respectively having significant difference(P<0.05). The SDF-1 expression level positively correlated with metastasis of lymph node and clinical stages(P<0.05),while no significant difference was observed in depth of invasion, grade of differentiation and the size of tumor.Conclusion: (1) E-Cadherin, CXCR4 and SDF-1 all have high relationship with the metastasis of lymph node. (2) E-Cadherin, CXCR4 and SDF-1 play an important role in development, invasion and metastasis in gastric cancer.
