Objective:To explore the effects and mechanism of atrovastatin on expression of the NF-κB on retionpathy of diabetic rats.Methods:A total of 42 male SD rats were randomly taken out 28 to make diabetes model by induced injection of 65mg streptozotocin (STZ)into enterocoelia,the rest was normal control group.After the model maded, atrovastatin(2mg/kg/d) were given the treated group,and normal control group and diabetic rats without treatment group was given equivalent water.After 3 and 6 months.the rats were killed.Isolated total RNA of the retina from one eye for each rat,and the other eye was prepared for the examination of immunohistochemistry. Determinated the NF-κB mRNA expression amoug three groups by the method of RT-PCR. Observed the distribution of the NF-κB in the retina, and compared the diference of three group.Results:PCR shows that NF-κB mRNA was increased in the retina of diabetic compared with control rats (P<0.05). Drugs treated animal showed significiantly decreased leveals of NF-κB mRNA in the retina compared with the untreated diabetic group (P< 0.05).IHC shows that there existed positive cells in the vascular layer.The increased number of the specific dyed cells in the diabetic retina also showed darer color (P<0.05).Additionaly,the drug treated retina trended to have fewer positive cells and lighter color compared with diabetic ones (P<0.05)Conclusion:Atovastatin can down-regulate the expression of NF-κB and suppress the increased levelof NF-κB protein in the retina of diabetic rats,slow the progress of retinopathy.
