Objectives:To determine the expression of proteins and mRNA of hPOTl and p53 in gastric carcinoma and investigate their significance in occurrence and development of gastric carcinoma,as well as their clinical significance and the possible mechanism of carcinogenesis caused by HP infection.Methods:The expression of hPOT1 and P53 proteins were detected in 53 gastric carcinoma specimens by SP immunohistochemical method;The expression of hPOT1mRNA and P53mRNA were detected in 53 gastric carcinoma specimens by in situ hybridization (ISH); and 10 normal gastric mucosa were control.HP infection was examed by using Warthin-Starry method in 53 gastric carcinoma specimens.Rusults:(1)In 53 cases of gastric carcinoma, the positive expression rate of hPOT1 protein was 84.91%(45/53),the positive expression of hPOTl protein was more frequently detected in gastric carcinoma than in 10 normal gastric mucosa 30%(3/10)(P<0.01).The positive expression of hPOT1 protein was significantly correlated with various histological type,differentiation and advanced degree of pathologic characteristics in gastric carcinoma(P<0.05),but not with age,sex, part, lvmph node metastasis(P>0.05).(2)In 53 cases of gastric carcinoma, the positive expression rate of p53 protein was 60.38%(32/53),the expression of p53 protein was more frequently detected in gastric carcinoma than in 10 normal gastric mucosa 10%(P<0.05).The positive expression of p53 protein was significantly correlated with advanced degree of pathologic characteristics in gastric carcinoma(P<0.05).but not with age,sex, part,lvmph node metastasis,various histological type and differentiation(P>0.05).(3)In 53 cases of gastric carcinoma,the positive expression rate of hPOT1mRNA was 58.49%(31/54),the expression of hPOT1mRNA was more frequently detected in gastric carcinoma than in 10 normal gastric mucosa 10%(1/10) (P<0.05).The positive expression of hPOT1mRNA was significantly correlated with advanced degree of pathologic characteristics in gastric carcinoma(P<0.05).but not with age,sex, part,lvmph node metastasis,various histological type and differentiation(P>0.05).(4)In 53 cases of gastric carcinoma,the positive expression rate of p53mRNA was 30.20%(16/53),the positive expression of p53mRNA was less frequently detected in gastric carcinoma than in 10 normal gastric mucosa 70%(7/10)(P<0.05).(5)The rate of HP infection in 53 cases of gastric carcinoma was 52.83%(28/53).(6)The positive expression of hPOT1 protein was positively corelated with the positive expression of p53 protein in gastric carcinoma (r=0.416,P<0.05). The positive expression of hPOTl protein was positively corelated with the positive expression of hPOTlmRNA in gastric carcinoma (r=0.394,P<0.05).The positive expression of p53 protein was negatively corelated with the positive expression of p53mRNA in gastric carcinoma (r=-0.476,P<0.05).(7)The positive expression rate of hPOT1 protein in gastric carcinoma group with HP infection was significantly higher than that in gastric carcinoma group without HP infection(P<0.05).The positive expression rate of p53 protein and hPOT1mRNA in gastric carcinoma group with HP infection was significantly higher than that in gastric carcinoma group without HP infection(P<0.01).Conclusions:(1)hPOT1 is associated with occurrence and development of gastric carcinoma,Detecting the expressions of hPOT1 protein and hPOT1mRNA plays an important role in predicting the prognosis of patients with gastric carcinoma.(2)mutp53 is associated with occurrence and development of gastric carcinoma;Loss of wtp53 plays an important role in with occurrence of gastric carcinoma.(3)hPOT1 and mutp53 are positively associated with occurrence and development of gastric carcinoma. (4) HP infection is associated with hPOTl and p53 in occurrence of gastric carcinoma.
