| Background and purposeAcute myocardial infarction (AMI) is one of the main diseases which threat human life and health , in particular for the elderly people. In recent years, apoptosis was received wide attention by scientists, who find that apoptosis takes part and plays an important role in the incidence of myocardial infarction, heart failure after myocardial infarction. caspase-3 is a key element in implementation of of apoptosis, and plays an important role in many ways about apoptotic signal transduction .Edaravone, a free radical scavenger, which has been used in clinical treatment of neurological diseases , was used in our reseach. And the rat model of myocardial infarction was established by ligation anterior descending artery. Myocardial infarct size was observed by TTC staining .The expression of caspase- 3 and GSH-Px also were analysed. The effect and application prospects of edaravone on the treatment of myocardial infarction also were studied.Methods60 SD rats were randomly divided into 3 groupsâ‘ sham group:normal saline was injected intravenously (4mg/kg) at 30 minutes before coronary artery surgery, no ligation of coronary artery.â‘¡acute myocardial infarction(AMI) group: Anterior descending coronary artery was ligated in AMI group.â‘¢edaravone treatment group: Anterior descending coronary artery was ligated, and edaravone was injected intravenously (4mg/kg) at 30 minutes before coronary artery surgery. At 24h after ligation, six rats in each group were stained by TTC (Triphenyi tetrazolium chloride) and myocardial infarct size was analysed, Serum GSH-Px activity was detected by the way of colorimetric Methods, mRNA of caspase-3 was detected by the way of RT-PCR. Results1. morphology changs under light microscope:Under light microscope, morphological characteristics of myocardial cells in sham group are normal. Necrosis was seen in myocardial infarction group, Around the necrosis, condensation and margination of chromatin were found in viable cells and nucleus are stained into dark.Necrosis was improved in Edaravone treated group.2. gravimetric determination in myocardial infarction:In myocardial infarction group, the proportion of myocardial infarct area with left ventricular and the whole heart weight were 24.78% and 15.2%; In edaravone group , infarct area reduced significantly, the proportion were 18.11% and 12.23% .3. The expression of caspase-3 :The expression level of caspase-3 was measured by quantitative RT-PCR. mRNA expression of caspase-3 in edaravone treatment group reduced apparently than that in AMI group. The lowest expression of caspase-3 was seen in Sham group.4. Serum GSH-Px:Serum GSH-Px in edaravone treatment group increased apparently than that in AMI group.ConclusionsPretreatment with edaravone can reduce infarct size significantly, the expression of caspase-3 also reduce , and serum GSH-Px increase. Protection of edaravone maybe have relationship with its roles of reduction of caspase-3 and increase of serum GSH-Px. |
