Objective:To investigate the effect of Enalapril on expressoin of integrinαV in rat liver fibrogenesis induced by carbon tetrachloride.Methods:Hepatic fibrosis models were induced by the method of injecting carbon tetrachloride(CCl4) into the rat abdominal cavity. Thirty-six males Wistar rats were randomly divided into three groups as follows: normal control group(n=8), model control group(n=14) and group treated by Enalapril(n=14). All rats were sacrificed at the end of experiment, and their blood and livers were collected for analysis.:the protein level of integrinαV in the liver were tested by RT-PCR;the expressing of IntegrinαV proteins were detected by immunohistochemistry staining;Serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were measured by biochemical analysis;Histological examination was also done.Results:1. Survival of rats: normal control group all survived ; model control group15,32,43,49,51,52 days in the death of one and 54 days in the death of two; group treated by Enalapril 37,40,52 days in the death of one.2. General condition: the normal control group of rats in good condition,color gloss, response, and weight gain; the model control group of rats appeared dark matte color, activity less, decreased food appetite , and eight rats died during the experiment .Compared with model control group, the group treated by Enalapril were better in shiny coat,weight and appetite, etc . This indicates that Enalapril may prevent the progression of the hepatic fibrosis and have an important cure role for hepatic fibrosis.3. Compared with the normal control group, the protein level of integrinαV in the model group was improved significantly as well as the blood serum ALT and AST. 4. Compared with the model group, the protein level of integrinαV and the blood serum ALT and AST were decreased obviously in the group treated by Enalapril. Conclusion:Enalapril can reduce the severity of liver fibrosis in rats by holding back integrinαV protein expression and inhibiting HSC activation, so Enalapril may be an effective anti-inflammation and anti-fibrosis drug.
