Objective To investigate the expression and clinical significance of TLR2,TLR4 in mature pregnancy women vaginal epithelial cells and placental tissues and to analyze the relationship between the expression of TLR2,TLR4 and that of pregnancy RVVC.Methods Immunohistochemical staining of SP was performed to determine the protein expressions of TLR2 and TLR4 in human vaginal epithelial cells and placental tissues of 30 mature pregnancy RVVC,30 mature pregnancy VVC and 30 Normal mature pregnancy. With the Motic Med digital medical image analysis system to measured the average optical density (OD) value. All date was analyzed using t test, variance analysis s by SPSS17.0 software(p<0.05).Results1 TLR2 expressed in vaginal epithelial cells in patients with Normal mature pregnancy, mature pregnancy VVC and mature pregnancy RVVC,and were expressed as 0.434±0.032, 0.490±0.094 and 0.375±0.062, the differences among the three groups was statistically Significance (F = 21.722 P <0.01). The expression of TLR2 in mature pregnancy VVC group's vaginal epithelial cells (0.490±0.094) was significantly higher than the normal mature pregnancy group (0.434±0.032) (P<0.05).The positive expression mature pregnancy RVVC group's vaginal epithelial cells(0.375±0.062) was significantly lower than the normal mature pregnancy group (0.434±0.032) (P <0.05).2 TLR4 expressed in vaginal epithelial cells in patients with Normal mature pregnancy, mature pregnancy VVC and mature pregnancy RVVC , and were expressed as 0.315±0.022, 0.372±0.039 and 0.245±0.038, the differences among the three groups was statistically Significance (F= 105.645 P <0.01). The expression of TLR4 in mature pregnancy VVC group's vaginal epithelial cells(0.372±0.039) was significantly higher than the normal mature pregnancy group(0.315±0.022)(P<0.05).The positive expression mature pregnancy RVVC group's vaginal epithelial cells(0.245±0.038)was significantly lower than the normal mature pregnancy group(0.315±0.022)(P <0.05).3 TLR2 expressed in placenta of patients'with Normal mature pregnancy, mature pregnancy VVC and mature pregnancy RVVC , and were expressed as 0.407±0.025,0.464±0.017 and 0.350±0.034, the differences among the three groups was statistically Significance (F= 65.573 P<0.05). The expression of TLR4 in mature pregnancy VVC group's placenta(0.464±0.017)was significantly higher than the normal mature pregnancy group(0.407±0.025)(P<0.05).The positive expression mature pregnancy RVVC group's vaginal epithelial cells(0.350±0.034)was significantly lower than the normal mature pregnancy group(0.407±0.025)(P <0.05).4 TLR4 expressed in placenta of patients'with Normal mature pregnancy, mature pregnancy VVC and mature pregnancy RVVC , and were expressed as 0.397±0.030,0.455±0.035 and0.332±0.033, the differences among the three groups was statistically Significance (F = 107.378 P <0.05). The expression of TLR4 in mature pregnancy VVC group's placenta(0.455±0.035)was significantly higher than the normal mature pregnancy group(0.397±0.030)(P <0.05).The positive expression mature pregnancy RVVC group's vaginal epithelial cells(0.332±0.033)was significantly lower than the normal mature pregnancy group(0.397±0.030)(P <0.05).5 Using t test compared the expression of TLR2 and TLR4 between the two groups in the vaginal epithelium, the positive expression level of TLR2 in the vaginal epithelium group (0.433±0.082)was higher than the placenta group (0.404±0.051), the difference was statistically Significance (P <0.05); the positive expression level of TLR4 in the placenta group (0.311±0.062)was higher than the vaginal epithelium group (0.405±0.065), the difference was statistically Significance (P <0.05). Conclusions:1 TLR2 and TLR4 expressed in vaginal epithelial cells and placenta of patients with Normal mature pregnancy, mature pregnancy VVCand mature pregnancy RVVC.2 Strong expression of TLR2 in the vaginal epithelium, and strong expression of TLR4 in the placenta.3 TLR2 and TLR4 may be involved in the occurrence and development of mature pregnancy VVC,and TLR2 and TLR4 may be affected its recurrence.4 The local immunity of Placenta may be involved in the procession which against Candida during mature pregnancy RVVC's infection.
