Objective: To research Lead acetate Exposure in SH—SY5Y Cells influence the expression of gene APP and DNMT1 and the global DNA methylation.Methods and results:1.Cultivate the SH -- SY5Y cells, and exposed them to different concentrations of lead acetate solution, using the MTT method to detect different doses of lead acetate effect SH—SY5Y cell growth curve .2.The DNA methyltransferase 1 (DNMT1) and the APP mRNA expression level were investigated. The results showed that the exposure of the two concentrations of Pb acetate could make the APP mRNA up regulation and the DNMT1 mRNA down regulation.3. The global methylation pattern was reduced by 6.8% and 10.6% after 7 days exposure; and the DNA methyltransferase 1 expression was reduced by 8.8% and 25.8% after 7 days exposure in this research. Conclusion:Lead acetate produce neurotoxicity by changing the APP and DNMT1 genes mRNA expression , its mechanisms may alter the Global DNA methylation level to increase the risk from Pb exposure that may promotes the pathogenesis of AD.
