Objective : Flavonoids, terpene saponins class,organic acids phenolic are effective Constituents in Triditional Chinese Medicine, three previous studies have shown that these compounds have extensive antiviral effect, this experiment is designed to screening the 18 kinds of effective component to find antiviral activity in guinea pig cytomegalovirus (GPCMV).Methods: In this test guinea pig embryos lung cell in the vitro as the research object, with acyclovir as tested positive control, Using four methyl azo thiazole salt colorimetric method (MTT) and cell pathological changes method (CPE) respectively detected Flavonoids (Baicalin, Baicalein, Luteoloside,Luteolin,Naringin,Naringenin,Apiolin,Rutin,Puerarin,Kaempferol),, organic acids phenolic (chlorogenicacid,coffeeacid,mangiferin), terpene saponins compounds oleanolic acid and ursolic acid,Saikosaponin B, Astragaloside IV,Dioscin) 18 Effective Constituents Of TCM anti-GPCMV effective,And with Ganciclovir as control.Results:①This result finds that Flavonoids (Baicalin, Baicalein, Luteoloside,Luteolin,Naringin,Naringenin,Rutin) and terpene saponins compounds(oleanolic acid,ursolic acid) have anti-GPCMV effective,first are Baicalein and Luteolin,second are Naringenin,oleanolic acid and ursolic acid ,third are Baicalin and Rutin,the last are Luteoloside and Naringin;②The Effective Constituents of cell cytotoxicity from high to low are respectively Naringenin,Baicalein,Luteolin,Luteoloside,Rutin,oleanolic acid,ursolic acid,Baicalin,Naringin③The TI of Effective Constituents from high to low are respectively, Baicalein,Luteolin,ursolic acid,oleanolic acid,Naringenin,Naringin,Luteoloside,Rutin④The Effective Constituents have better anti-GPCMV effective than Ganciclovir and Jinye baidu granules,but the cell cytotoxicity more than that of Ganciclovir and Jinye baidu granules, then,The TI Baicalein,Luteolin,ursolic aci,oleanolic acid is higher than Ganciclovir.Conclusion: The Effective Constituents such as Baicalein are expected to become new hot Chinese medicine for resistance HCMV. Objective:According to the first part results found that The Baicalein, luteolin, ursolic acid, oleanolic acid have anti-GPCMV effective,more than Ganciclovir, This study section is designed to observe The Baicalein, luteolin, ursolic acid, oleanolic acid have or not influence in virus adsorption, penetration, reproduction in each stage .Methods: Using the method of synchronous infections observed The Baicalein, luteolin, ursolic acid, oleanolic acid mainly play a role.in the what stage:virus adsorption, penetration, reproduction stage . Through the application of RT - PCR method, fluorescence quantitative polymerase chain reaction (PCR) method to Observe the Baicalein, luteolin and Ganciclovir is whether have any effects in GPCMV GP129 mRNA level . Results:①The Baicalein, luteolin, ursolic acid all can affect viral replication process, also the Baicalein can affect penetration stage,but the other two are not on the adsorption and penetration, Oleanolic acid have no influence on three stages;②The Baicalein, luteolin have effects in GPCMV GP129mRNA level respectively,while are similar with Ganciclovir. Conclusion: The Baicalein, luteolin, ursolic acid can affect viral replication process, the replication of influentialof The Baicalein luteolin and and Ganciclovir more revlant to GP129 expression .
