Objective The role of high mobility group box 1 in hepatocellular carcinoma is unknown. We detecte the expression differences of HMGB1 between HCC and lateral carcinoma hepatic tissue , and evaluate contributions of high mobility group box 1 in hepatocellular carcinoma.Methods Immunohistochemisty and Western blot analysis were used to detect the expression of HMGB1 between HCC and lateral carcinoma hepatic tissue. Then, the expression difference and clinicopathologic significance of HMGB1 were analyzed.Results The expression of HMGB1 were detected in the nucleus at HCC,lateral carcinoma hepatic tissue and normal hepatic tissue, but the positive ratio of HMGB1 levels in cytoplasm of lateral carcinoma hepatic tissue (61.76%)was significantly higher than in HCC (8.82%), and positive correlation were found between HMGB1 in cytoplasm of lateral carcinoma hepatic tissue and AFP (P < 0.05), and between HMGB1 in cytoplasm of lateral carcinoma hepatic tissue and the size of tumour (P< 0.05). The expression of HMGB1 detected by western-blot was present in both HCC and lateral carcinoma hepatic tissue at similar levels.Conclusions the positive ratio of HMGB1 levels in cytoplasm of lateral carcinoma hepatic tissue was significantly higher than in HCC, and this evidence may indicate that the cells of lateral carcinoma hepatic tissue release HMGB1 to extracellular, and may contribute to the tumor progression of HCC.
